褪黑素受体基因多态性与卵巢储备功能相关性的研究

摘　　要：目的探讨褪黑素受体基因多态性与卵巢储备功能的相关性。方法选取助孕的患者158例为研究对象,其中卵巢储备功能减退组40例,正常卵巢储备组118例,采用测序法检测褪黑素受体1B基因rs10830963位点基因型,采用酶联免疫吸附试验测定血清抗缪勒管激素及褪黑素水平,比较卵巢储备功能减退组(DOR组)与正常卵巢储备组(NOR组)患者褪黑素受体1B基因rs10830963位点基因型及等位基因分布特点;比较不同基因型患者卵巢储备相关指标的表达水平。结果褪黑素受体1B基因rs10830963位点有GG、GC、CC 3种基因型,2组患者均以GC基因型表达为主,但卵巢储备功能减退组患者中GG基因型比例较高,且差异有统计学意义(P<0.05);2组患者等位基因分布频率差异无统计学意义(P>0.05);GG基因型组、GC基因型组及CC基因型组中窦卵泡数分别为(8.93±5.71)个、(12.97±5.87)个、(12.27±6.04)个,GG基因型与GC基因型及CC基因型相比AFC水平较低,且差异有统计学意义(P<0.05),而GC基因型与CC基因型间AFC差异无统计学意义(P>0.05)。结论褪黑素受体1B基因rs10830963位点基因多态性可能与卵巢储备功能具有相关性。Objective To investigate the correlation between melatonin receptor gene polymorphism and ovarian reserve function.Methods A total of 158 patients were selected in the study and the patients including 40 patients in the diminished ovarian reserve group and 118 patients in the normal ovarian reserve group.The rs10830963 genotype of melatonin receptor 1B gene was detected by sequencing,and the serum anti-Mullerian hormone and melatonin levels were determined by enzyme-linked immunosorbent assay(ELISA).The genotype and allele distribution of rs10830963 locus of melatonin receptor 1B gene in patients with diminished ovarian reserve group and normal ovarian reserve group were compared.The expression levels of ovarian reserve related indexes in patients with different genotypes were compared.Results There were GG,GC and CC genotypes at rs10830963 of melatonin receptor 1B gene,GC genotype expression was predominant in both groups.However,the proportion of GG genotype was higher in patients with diminished ovarian reserve group than normal ovarian reserve group(P<0.05).There was no significant difference in allele frequency between the two groups(P>0.05).The number of antral follicle count in GG genotype,GC genotype and CC genotype were respectively 8.93±5.71,12.97±5.87 and 12.27±6.04.GG genotype had lower AFC level than GC genotype and CC genotype,and the difference was statistically significant(P<0.05).There was no significant difference in AFC between GC genotype and CC genotype(P>0.05).Conclusion The s10830963 polymorphism of melatonin receptor 1B gene may be associated with ovarian reserve function.
