摘  要:目的 对倒心盾翅藤Aspidopterys obcordate藤茎的化学成分进行研究。方法 采用正相硅胶、反相ODS、MCI gel CHP 20P等柱色谱及制备高效液相色谱等分离方法进行分离纯化,运用旋光、核磁共振、质谱和圆二色光谱等多种波谱学技术鉴定化合物的化学结构;并采用一水草酸钙结晶(COM)诱导的大鼠肾小管上皮NRK-52E细胞损伤模型,对分离得到的单体成分进行活性筛选,初步探讨各单体成分对COM致NRK-52E细胞损伤的保护作用。结果 从倒心盾翅藤藤茎70%乙醇提取物中分离得到17个化合物,分别鉴定为槲皮素(1)、(+)-儿茶素(2)、异黄烷香豆素(3)、龙脑苷(4)、大柱香波龙烷-9-O-β-D-葡萄糖苷(5)、长春花糖苷Ⅱ(6)、2,3-seco-sonderianol(7)、木栓酮(8)、2α,3β,23-三羟基-12-烯-28-齐墩果酸(9)、(24S)-24-乙基胆甾-3β,5α,6β-三醇(10)、辛二酸单甲酯(11)、1-壬二酸甘油酯(12)、(9S,10E,12S,13S)-9,12,13-三羟基-10-十八碳烯酸(13)、(9S,10E,12S,13S,15Z)-三羟基十八烷-10,15-二烯酸(14)、异丙醇-[6-O-(β-D-木糖基)]-β-D-吡喃葡萄糖苷(15)、3,4-二羟基苯甲酰胺(16)和4’-O-(β-D-吡喃葡萄糖基)-维生素B5(17)。结论 化合物3~5、7、9~17共13个化合物为首次从盾翅藤属中分离得到;并首次报道了化合物17的13C-NMR谱数据。其中,化合物1~3、10、13和14在不影响细胞活力的情况下,对COM所致NRK-52E细胞的损伤具有明显的抑制作用。Objective To study the chemical constituents of the 70% Et OH extract of Aspidopterys obcordate vines.Methods Silica gel,ODS,and MCI gel CHP 20P column chromatographies,as well as preparative high-performance liquid chromatography were applied for the constituents' isolation from the 70% Et OH extract.Then,[α]D,NMR,MS,CD,and other related spectroscopic analyses were used to identify their structures.Moreover,the protective effects of each purified compound on calcium oxalate monohydrate(COM) induced rat renal tubular epithelial cells(NRK-52E cells) injury were investigated.Results A total of 17 compounds,quercetin(1),(+)-catechin(2),isoflavanocoumarin(3),1-borneol-β-D-apisyl-β-D-glucopyranoside(4),lauroside B(5),roseoside II(6),2,3-seco-sonderianol(7),friedelin(8),2α,3β,23-trihydroxyolean-12-en-28-oic acid(9),(24S)-24-ethylcholesta-3β,5α,6β-triol(10),suberic acid monomethyl ester(11),1-azeloyl-rac-glycer(12),(9S,10E,12S,13S)-9,12,13-trihydroxy-10-octadecenoic acid(13),fulgidic acid(14),2-[6-O-(β-D-xylopyranosyl)-β-D-glucopyranosyloxy]propane(15),3,4-dihydroxybenzamide(16),and 4'-O-(β-D-glucopyranosyl)-DL-pantothenic acid(17) were obtained from the 70% Et OH extract of A.obcordate vines.Conclusion Thirteen compounds including 3—5,7,and 9—17 were isolated firstly from Aspidopterys genus.Meanwhile,the 13C-NMR data of compound 17 was reported for the first time.And compounds 1—3,10,13,and 14 could significantly improve the survival rate of COM-induced NRK-52E cells injury,suggesting that they had some protective effect against the cells damage.
