摘  要:为探究霸王(Zygophyllum xanthoxylum)液泡膜Na^(+)区域化功能基因ZxNHX1和H^(+)-焦磷酸酶(H^(+)-pyrophosphatase)编码基因ZxVP1-1及角质层蜡质转运基因ZxABCG11对“新疆大叶”紫花苜蓿(Medicago sativa)生长发育及饲用品质的影响,测定野生型及ZxNHX1-VP1-1和ZxABCG11转基因紫花苜蓿在田间条件下的生长发育和饲用品质相关指标.结果表明,ZxNHX1-VP1-1转基因苜蓿的叶面积和干草产量分别比野生型高56.4%和74.5%,净光合速率和水分利用率均显著高于野生型(P<0.05),花期比野生型延长19d;ZxABCG11转基因苜蓿的叶面积和茎粗均显著大于野生型(P<0.05),净光合速率和水分利用率分别比野生型高81.7%和80.5%,花期比野生型延长38d;ZxNHX1-VP1-1转基因苜蓿的中性洗涤纤维、酸性洗涤纤维和酸性洗涤木质素含量显著低于野生型(P<0.05),相对饲用价值比野生型提高了35.4%.霸王ZxNHX1和ZxVP1-1的超表达提高了紫花苜蓿的生物量和饲用品质,并延长了开花时间;ZxABCG11的超表达对紫花苜蓿叶面积、茎粗和光合能力产生了影响,花期相比野生型有所延长.This study aimed to explore the effects of tonoplast Na^(+)compartmentalization gene ZxNHXI,H-PPase(H^(+)-pyrophosphatase)encoding gene ZxVP1-1 and cuticular wax transport gene ZxABCG11 from Zygophyllum xanthoxylum on the growth,development and forage quality of alfalfa(Xinjiang daye).We measured the indicators of growth,development and forage quality of wild type,ZxNHXI-VP1-1 and ZxABCG11 transgenic alfalfa under field conditions and the results showed that leaf area and hay yield of ZxNHXI-VP1-1 transgenic alfalfa were 56.4%and 74.5%higher than those of the wild type,respectively.The net photosynthesis rate and water use efficiency were significantly higher than those of the wild type(P<0.05).The flowering period was 19 days longer than that of the wild type.Leaf area and stem diameter of ZxABCG11 transgenic alfalfa were significantly increased compared to wild type(P<0.05).The net photosynthesis rate and water use efficiency were 81.7%and 80.5%higher than those of the wild type,respectively.The flowering period was 38 days longer than that of the wild type.Neutral detergent fiber,acid detergent fiber and acid detergent lignin of ZxNHXI-VP1-1 transgenic alfalfa were significantly lower than those of the wild type(P<0.05).On the contrary,the relative feed value of ZxNHX1-VP1-1 transgenic alfalfa was 35.4%higher than that of the wild type.These results showed that the overexpression of ZxNHXI and ZxVP/-1 increased the biomass and nutritional quality,and prolonged flowering time of alfalfa.Overexpression of ZxABCG11 affected the leaf area,stem diameter and photosynthetic capacity of alfalfa.The flowering period of ZxABCG11 transgenic alfalfa was longer than that ofwild type.
