摘 要:为了探讨母鼠长期铜暴露对子代大鼠血清和睾丸睾酮水平以及睾丸细胞凋亡的影响,本试验选取24只胎次相同、体重相近[(44.04±3.05)g]的健康雌性SD大鼠,随机分为3个组,每组8只,分别为A组(基础日粮)、B组(基础日粮+CuSO_(4)120 mg/kg)、C组(基础日粮+CuSO_(4)240 mg/kg),连续饲喂8周,与健康半同胞雄性SD大鼠配种,并在妊娠期和哺乳期持续饲喂相应组别日粮,子代断奶后均饲喂基础日粮至10周龄,在每组子代中随机选取8只雄性大鼠,分别标记为A0、B0和C0组,测定血清和睾丸中睾酮水平以及睾丸细胞凋亡相关因子的mRNA和蛋白相对表达水平。结果显示,随着亲代日粮铜含量提高,与A0组相比较:(1)C0组中血清和睾丸的睾酮水平显著降低(P<0.05);(2)B0组和C0组中凋亡因子Bax mRNA和蛋白相对表达水平均显著增加(P<0.05)、Caspase-3 mRNA相对表达水平显著增加(P<0.05),而其蛋白相对表达水平仅在C0组显著增加(P<0.05);(3)C0组中抗凋亡因子Bcl-2 mRNA和蛋白相对表达水平显著降低(P<0.05)。结果表明,母鼠长期高浓度铜暴露会影响雄性子代大鼠睾酮合成,引起子代大鼠睾丸细胞凋亡。In order to assess the effects of long-term copper exposure in maternal rats on testosterone levels of serum and testis and testicular cell apoptosis in offspring rats,this study randomly allocated 24 healthy female SD rats with same parity and similar body weight[(44.04±3.05)g]into 3 groups,with 8 rats in each group.The experimental groups are as follows:group A(basal diet),group B(basal diet+CuSO_(4)120 mg/kg)and group C(basal diet+CuSO_(4)240 mg/kg).After 8 weeks of continuous feeding,female rats were mated with healthy half-sibling male SD rats and continued feeding diets with different copper levels in pregnancy and lactation.The progenies were fed basal diet to 10 weeks of age after weaning.In each progeny group,8 male rats were randomly selected and labeled as A0,B0 and C0 groups,respectively.The testosterone levels in serum and testis and relative expression levels of testicular apoptotic genes and proteins were determined.The results showed that with increased copper content in parental diet,compared with A0 group:(1)the serum and testicular testosterone levels were significantly decreased in C0 group(P<0.05);(2)in B0 group and C0 group,the relative expressions of Bax mRNA and protein were significantly increased(P<0.05)and the relative expressions of Caspase-3 mRNA were significantly increased(P<0.05),while the relative expressions of Caspase-3 protein was significantly increased only in C0 group(P<0.05);(3)the relative expressions of anti-apoptotic factor Bcl-2 mRNA and protein were significantly decreased in C0 group(P<0.05).Thus,long-term exposure to high concentrations of copper in female rats affects testosterone synthesis and apoptosis of testicular cells in male offspring rats.
