摘 要:目的 构建适宜的黄柏提取工艺操作空间,优化黄柏水提工艺。方法 以小檗碱提取率、黄柏碱提取率、绿原酸提取率、干膏率为关键质量属性,运用鱼骨图结合Plackett-Burman设计筛选出提取时间、提取温度和提取次数为关键工艺参数。通过Box-Behnken设计建立工艺统计模型,分别采用指标叠加法和Monte Carlo法两种设计空间算法多目标综合比较,建立提取工艺的设计空间并对不同达标概率进行验证。结果 在达标概率为0.9条件下最终确定黄柏提取工艺操作空间为提取2次,温度为87~95℃,时间为70~100 min。结论 在构建的工艺参数设计空间内黄柏提取工艺品质稳定。Objective To optimize the water stripping process of Phellodendri Chinensis Cortex,and construct the process design space by two multi-objective algorithms.Methods The extraction rate of berberine,xanthine,chlorogenic acid and dry paste were taken as the key quality attributes,and the extraction time,extraction temperature and extraction times were screened out by fish bone diagram combined with PlackettBurman design.The mathematical model was established by Box-Behnken design,and the multi-objective comprehensive comparison of two design space algorithms,index superposition method and Monte Carlo method was used to establish the design space of the extraction process and verify the different probability of achieving the standard.Results Under the condition that the probability of reaching the standard was 0.9,the operation space of the extraction process was finally determined to be extracted twice,the temperature was 87-95 ℃,and the time was 70-100 min.Conclusion Operation in the design space of process parameters can ensure the stable quality of the extraction process of Phellodendrin Chinense Cortex.
