夜间温度对栽培膜荚黄芪生长、药用成分及相关基因表达的影响

摘  要:以膜荚黄芪(Astragalus membranaceus)为试材,采用盆栽试验法,设置5个夜间温度(4、8、12、16、20℃),探索了夜间不同温度对黄芪生长、药用成分及相关基因表达的影响,以期为膜荚黄芪的夜间管理提供参考依据。结果表明:夜间温度对黄芪的生长发育具有显著影响。随着温度升高,黄芪生长参数、SPAD值及气孔导度整体呈先升高后降低趋势,以16℃效果整体最佳,其显著促进了黄芪根系发育、地上部生长,增加了干物质质量、SPAD值和气孔导度,并提高了初级代谢物(可溶性糖、淀粉)和皂苷、总酚及异黄酮相关组分等次级代谢物含量。基因表达分析表明,当夜间温度为16℃时,颗粒结合淀粉合酶基因(AmGBSS、ClGBSS)、核酮糖二磷酸羧化酶基因(AmRBCL)和铁氧还蛋白(ClFDX)显著上调表达。综上,16℃是栽培黄芪生长发育和品质形成的最佳夜间温度。A pot experiment took Astragalus membranaceus as the test material,five nighttime temperatures(4,8,12,16,20 C)were set up to explore the effects of different nighttime temperatures on the growth,medicinal components and related gene expression of Astragalus membranaceus,in order to provide reference for the night management of Astragalus membranaceus.The results showed that nighttime temperature had a significant effect on the growth and development of Astragalus membranaceus.SPAD value and stomatal conductance of Astragalus membranaceus increased first and then decreased,and the effect of 16 C was the best.It significantly promoted the root development and shoot growth of Astragalus membranaceus,increased dry matter weight,SPAD value and stomatal conductance.Furthermore,the nighttime temperature of 16℃increased the content of primary metabolites(soluble sugars,starch)and secondary metabolites such as saponins,total phenols and isoflavones.Gene expression analysis showed that the expression of granule-bound starch synthase(AmGBSS),ribulose diphosphate carboxylase(AmRBCL,CIGBSS)and ferredoxin(CIFDX)was significantly up-regulated when the nighttime temperature was 16 C.In conclusion,16℃C was the best nighttime temperature for the growth,development and quality of Astragalus membranaceus.
