摘  要:目的探讨去亚甲基小檗碱(DMB)对1-甲基-4-苯基吡啶离子(MPP+)诱导的SH-SY5Y细胞线粒体caspase依赖性凋亡通路的影响。方法培养SH-SY5Y细胞,将其分为对照组、MPP+组、小檗碱(BBR,DMB的前体)+MPP+组、司莱吉兰(selegiline,与DMB同为单胺氧化酶B抑制剂)+MPP+组、DMB+MPP+组。应用免疫印迹法检测各组bcl-2/bax比值以及cleaved caspase-3蛋白的表达。结果与对照组相比,MPP+组bcl-2/bax比值明显降低(F=42.58,q=17.89,P<0.001),DMB、BBR、selegiline处理均可以抑制MPP+诱导的bcl-2/bax比值降低(q=6.71~10.63,P<0.001),且selegiline处理效果强于BBR和DMB(q=3.93、3.59,P<0.05)。与对照组相比,MPP+组cleaved caspase-3蛋白表达明显上调(F=111.60,q=29.44,P<0.001),DMB、BBR、selegiline处理可以降低MPP+诱导的cleaved caspase-3蛋白表达上调(q=10.74~13.53,P<0.001),且3种药物处理效果差异无显著性。结论对于MPP+诱导的SH-SY5Y细胞,DMB可以升高bcl-2/bax比值,降低cleaved caspase-3蛋白的表达。Objective To investigate the effects of demethyleneberberine(DMB)on the mitochondrial caspase-dependent apoptosis pathway in SH-SY5Y cells treated with 1-methyl-4-phenylpyridinium(MPP+).Methods SH-SY5Y cells were cultured and divided into control group,MPP+group,berberine(BBR,the precursor of DMB)+MPP+group,selegiline(a monoamine oxidase B inhibitor similar to DMB)+MPP+group,and DMB+MPP+group.The ratio of bcl-2/bax and the expression of cleaved caspase-3 protein were determined by Western blot for each group.Results Compared with that in the control group,the bcl-2/bax ratio in the MPP+group was significantly lower(F=42.58,q=17.89,P<0.001).DMB,BBR,and selegiline all significantly inhibited the MPP+-induced decrease in the bcl-2/bax ratio(q=6.71-10.63,P<0.001),and the effect with selegiline was significantly greater than that with BBR(q=3.93,P<0.05)and that with DMB(q=3.59,P<0.05).Compared with the control group,the MPP+group showed significantly higher expression of cleaved caspase-3 protein(F=111.60,q=29.44,P<0.001).DMB,BBR,and selegiline significantly reduced MPP+-induced upregulation of cleaved caspase-3 protein(q=10.74-13.53,P<0.001),with no significant difference in the treatment effects of the three drugs(P>0.05).Conclusion For SH-SY5Y cells induced by MPP+,DMB can increase the ratio of bcl-2/bax and decrease the expression of cleaved caspase-3 protein.
