摘 要:【目的】探明冬青卫矛响应低温的生理机制,为其引种和培育提供参考。【方法】以冬青卫矛(Euonymus japonicus)一年生扦插苗为材料,经不同人工低温胁迫24 h后,测定叶片的叶绿素含量及荧光参数、相对含水量、相对电导率、渗透调节物质含量、抗氧化酶活性等,复温处理后观测生长表现。【结果】随温度降低,叶绿素含量、PSⅡ最大光化学量子产量和相对含水量呈下降趋势;相对电导率、可溶性糖和可溶性蛋白含量、过氧化物酶活性呈上升趋势;光化学猝灭系数、脯氨酸含量、超氧化物歧化酶和过氧化氢酶活性先上升后降低;非光化学猝灭系数为“降-升-降”变化趋势;温度与各生理指标之间及绝大多数生理指标之间显著相关。多项生理指标在0℃和-4℃处理期间变化幅度小或不显著,植株能恢复生长,而在-8℃和-12℃处理下变化幅度大甚至出现转折,植株不能恢复生长。【结论】人工低温胁迫下,冬青卫矛一年生扦插苗的抗寒反应是不同生理过程综合作用的结果,其可以较好地耐受-4℃以上的低温胁迫,但无法抵御-8℃以下低温。[Objective]This study aims to explore the physiological mechanisms of E.japonicus in response to low temperature stress,and provides reference for introduction and cultivation.[Methods]The annual cuttings of Euonymus japonicus were used as materials,and after treatment with different gradients of artificial low temperature stress,the chlorophyll contents,fluorescence parameters,relative water content,relative conductivity,osmotic adjustment substance contents and antioxidant enzyme activities of the leaves were measured.The growth performance was observed after rewarming treatment.[Results]With the decrease of stress temperature,chlorophyll content,F v/F m and relative water content showed downward trends;relative conductivity,soluble sugar and soluble protein contents,and peroxidase activity showed upward trends;photochemical quenching coefficients,proline content,superoxide dismutase activity and catalase activity first increased and then decreased;the non-photochemical quenching coefficient showed a trend of"decrease-rise-decrease";temperature was significantly correlated with various physiological indicators,and most of physiological indicators were also significantly correlated.Compared with CK,most of the physiological indicators changed little or non-significantly at 0℃and-4℃,and the plants could resume growth;at-8℃and-12℃,the variation range was large or even turned,and the plants could not resume growth.[Conclusion]The low temperature resistance of the annual cuttings of E.japonicus was the result of the combined effect of different physiological processes,which can better tolerate the low temperature stress above-4℃,but cannot withstand the low temperature below-8℃.
