摘 要:目的探讨高海拔低氧诱导对C57BL/6小鼠肺组织细胞自噬及凋亡的影响。方法选取8周龄C57BL/6小鼠30只,分别饲养于西安市雁塔区(对照组)及青海省果洛藏族自治州(高原低氧组),连续饲养21 d后处死并采集肺脏组织。透射电镜下观察肺组织显微结构,记录自噬体数量;免疫组化法检测肺组织自噬相关因子Beclin-1、LC3B、p62以及凋亡相关因子Bax、Bcl-2阳性信号表达情况,Western blotting法及实时荧光定量PCR法检测Beclin-1、LC3B、p62、Bax、Bcl-2蛋白及mRNA表达。结果高原低氧组与对照组自噬体数量分别为(4.80±0.84)、(1.80±0.45)个,两组比较P<0.05。与对照组比较,高原低氧组肺组织Beclin-1、Bcl-2蛋白阳性信号表达增强,p62、Bax蛋白阳性信号表达减弱(P均<0.05),LC3B蛋白阳性信号表达无明显变化(P>0.05)。与对照组比较,高原低氧组肺组织Beclin-1蛋白相对表达量及LC3BⅡ/Ⅰ、Bcl-2/Bax均升高,p62蛋白相对表达量降低(P均<0.05)。与对照组比较,高原低氧组肺组织Beclin-1、LC3B mRNA相对表达量及Bcl-2 mRNA/Bax mRNA均升高,p62 mRNA相对表达量降低(P均<0.05)。结论高原低氧环境下的小鼠肺组织自噬及抗凋亡能力均存在一定程度的增强。Objective To investigate the effects of high altitude hypoxia on autophagy and apoptosis in lung tissues of C57BL/6 mice.Methods Thirty 8-week-old C57BL/6 mice were selected and housed in Yanta District,Xi'an City(control group)and Goluo Tibetan Autonomous Prefecture,Qinghai Province(plateau hypoxia group),respectively,and they were executed and their lung tissues were collected after 21 d of continuous feeding.The microstructure of lung tissue was observed under transmission electron microscopy and the number of autophagosomes was recorded;the expression lev⁃els of autophagy and apoptosis factors Beclin-1,LC3B,p62,Bax and Bcl-2 positive signals in the lung tissues were detect⁃ed by immunohistochemistry,and Beclin-1,LC3B,p62,Bax and Bcl-2 protein and mRNA expression levels were detect⁃ed by Western blotting and real-time fluorescence quantitative PCR.Results The number of autophagosomes in the pla⁃teau hypoxia group and the control group were 4.80±0.84 and 1.80±0.45,respectively,with statistically significant dif⁃ference between the two groups(P<0.05).Compared with the control group,the expression of Beclin-1 and Bcl-2 protein positive signals in the lung tissues of the plateau hypoxia group was enhanced,and the expression of p62 and Bax positive signals was diminished(both P<0.05),and there was no significant change in LC3B positive signal expression(P>0.05).Compared with the control group,the relative expression of Beclin-1 protein,LC3BⅡ/Ⅰand Bcl-2/Bax in⁃creased and the relative expression of p62 protein decreased in the plateau hypoxia group(all P<0.05).Compared with the control group,the relative expression of Beclin-1,LC3B and Bcl-2/Bax mRNA in the lung tissue of the plateau hypox⁃ia group increased,and the relative expression of p62 mRNA decreased(both P<0.05).Conclusions Autophagy and anti-apoptotic capacity of mouse lung tissues were enhanced to a certain extent in the high altitude hypoxic environment.
