摘  要:目的:探讨电针对帕金森病(PD)小鼠肠道ERK MAPK/NF-κB通路的影响。方法:采用鱼藤酮连续灌胃28 d构造PD小鼠模型,将C57BL/6小鼠随机分为对照组、模型组与电针组。电针组小鼠电针刺激“风府”“太冲”“足三里”穴,30 min/次,1次/d,连续治疗14 d。观察各组小鼠行为学变化;免疫组化法检测黑质及结肠组织中α突触核蛋白(α-synuclein,α-syn)的平均光密度;Western blot法检测结肠组织中目的蛋白表达;实时荧光定量PCR检测结肠组织中TNF-α和IL-1βmRNA的相对含量。结果:与对照组比较,模型组小鼠运动总路程、运动时间、穿过总分区及平均速度均降低,差异具有统计学意义(P<0.01);黑质和结肠组织中α-syn平均光密度升高,差异具有统计学意义(P<0.01);结肠组织中p-ERK MAPK、p-NF-κB蛋白表达及TNF-α和IL-1βmRNA水平显著升高,差异具有统计学意义(P<0.01)。与模型组比较,电针组小鼠运动总路程、运动时间、穿过总分区及平均速度均增加,差异具有统计学意义(P<0.01);黑质和结肠组织中α-syn平均光密度降低,差异具有统计学意义(P<0.01);结肠组织中p-ERK MAPK、p-NF-κB蛋白表达及TNF-α和IL-1βmRNA水平降低,差异具有统计学意义(P<0.01)。结论:电针“风府”“太冲”“足三里”可以改善PD小鼠的运动功能障碍,降低α-syn表达,同时抑制肠道ERK MAPK/NF-κB通路激活。Objective:To investigate the effect of electro-acupuncture(EA)on intestinal ERK MAPK/NF-κB pathway in mice with Parkinson's disease(PD).Methods:PD mice model was constructed by continuous gavage of rotenone for 28 d.C57BL/6 mice were randomly divided into the control group,the model group and the EA group.The EA group was treated with electro-needling Fengfu(DU16),Taichong(LR3)and Zusanli(ST36),30 min/time,once/day for 14 d.The behavioral changes of the mice were observed in each group.The mean optical density ofα-synuclein(α-syn)in substantia nigra and colon tissues was measured by immunohistochemistry.The expressions of target proteins in colon tissues were detected by Western blot method.The relative mRNA expressions of TNF-αand IL-1βin colon tissues were measured by real-time fluorescence quantitative PCR.Results:Compared with those in the control group,the total exercise distance,exercise time,crossing the total division and average speed of mice were all reduced(P<0.01);the mean optical density ofα-syn in nigrostriatal and colonic tissues was increased(P<0.01);the protein expressions of p-ERK MAPK and p-NF-κB,as well as the mRNA levels of TNF-αand IL-1βin colon tissues were significantly increased in the model group(P<0.01).Compared with those in the model group,the total exercise distance,exercise time,crossing the total division and average speed were increased(P<0.01);the mean optical density ofα-syn was decreased in nigrostriatal and colonic tissues(P<0.01);the protein expressions of p-ERK MAPK and p-NF-κB,as well as the mRNA levels of TNF-αand IL-1βin colonic tissues were decreased in the EA group(P<0.01).Conclusion:Electro-needling DU16,LR3 and ST36 can improve motor dysfunction,reduceα-syn expression and inhibit intestinal ERK MAPK/NF-κB pathway activation in PD mice.
