优降脂组方对ApoE基因敲除动脉粥样硬化模型小鼠的动脉粥样硬化相关细胞因子的影响

摘　　要：目的探讨优降脂组方对ApoE基因敲除(ApoE-/-)动脉粥样硬化模型小鼠的动脉粥样硬化相关细胞因子的影响。方法选择ApoE-/-小鼠45只,同时选择同遗传背景C57BL/6J小鼠雄性15只。造模成功后分为4组:模型组、正常对照组、优降脂组方实验组、阿托伐他汀对照组。实验结束后,测定4组小鼠血脂水平及动脉粥样硬化相关细胞因子水平。结果模型组小鼠TC、TG、LDL-C均高于正常对照组,HDL-C低于正常对照组,差异有统计学意义(P<0.05)。优降脂组方实验组和阿托伐他汀对照组TC、TG、LDL-C均低于模型组,HDL-C均高于模型组,差异有统计学意义(P<0.05)。模型组小鼠hs-CRP、TNF-α、MDA均高于正常对照组,NO低于正常对照组,差异有统计学意义(P<0.05)。优降脂组方实验组和阿托伐他汀对照组hs-CRP、TNF-α、MDA均低于模型组,NO均高于模型组,差异有统计学意义(P<0.05)。结论优降脂组方有助于降低血脂水平,对动脉粥样硬化发生和发展有一定抑制作用,为临床提供一定的实验理论依据。Objective To explore the effect of Youjiangzhi recipe on atherosclerosis related cytokines in ApoE gene knockout(ApoE-/-)atherosclerosis model mice.Methods 45 ApoE-/-mice were selected,while 15 male C57BL/6J mice with the same genetic background were selected.After successful modeling,they were divided into four groups:model group,normal control group,experimental group of Youjiangzhi recipe,and atorvastatin control group.After the experiment,the levels of blood lipid and atherosclerosis related cytokines in the four groups of mice were measured.Results The TC,TG,and LDL-C in the model group were higher than those of the normal control group,while the HDL-C of the model group was lower than that of the normal control group,there were statistically significant differences(P<0.05).The TC,TG,and LDL-C levels in the experimental group of the Youjiangzhi recipe and the atorvastatin control group were lower than those in the model group,while the HDL-C levels in the experimental group of the Youjiangzhi recipe and the atorvastatin control group were higher than those in the model group,there were statistically significant difference(P<0.05).Hs-CRP,TNF-α,MDA levels of the model group were higher than those in the normal control group,while NO levels in the model group were lower than those in the normal control group,there were statistically significant differences(P<0.05).In the experimental group of Youjiangzhi recipe and the atorvastatin control group,hs-CRP and TNF-α,MDA were lower than those in the model group,while NO was higher in the experimental group of the Youjiangzhi recipe and the atorvastatin control group than that in the model group,there were statistically significant differences(P<0.05).Conclusion Youjiangzhi recipe can help to reduce the level of blood lipid,inhibit the occurrence and development of atherosclerosis,and provide some experimental theoretical basis for clinical practice.
