摘 要:本试验对山东某兔场疑似感染兔瘟致死的病料进行中和试验、血凝和血凝抑制试验、PCR鉴定及测序比对。试验结果表明,4份分离的病毒上清液都能与1%人O型红细胞发生凝集,凝集效价均为1:1024;与兔病毒性出血症阳性血清中和反应后,对人O型红细胞没有凝集反应;PCR扩增结果显示4份送检样品均在286 bp位置出现了目的条带,经测序比对,该序列与中国的03/China/2016毒株VP60基因(登入号为MF669712.1)序列同源性最高,相似度为99.65%。试验结果充分说明该病毒为兔病毒性出血症病毒,该兔场流行疫病为兔瘟。In this study,neutralization test,hemagglutination and hemagglutination inhibition test,PCR identification and sequencing were carried out on the pathogenic materials suspected to have died from rabbit plague infection in a rabbit farm in Shandong province.The results showed that the 4 isolates of virus supernatant could agglutinate with 1%of human O-type erythrocytes,and the agglutination titer was 1:1024.There was no agglutination reaction to human O erythrocyte after neutralizing with rabbit viral hemorrhagic positive serum.The PCR amplification results showed that the target band appeared in the 286 bp position of the 4 samples.After sequencing,the sequence had the highest homology with the Chinese 03/China/2016 strain VP60 gene(login number:MF669712.1),and the similarity was 99.65%.The results of this experiment fully indicated that the virus was rabbit viral hemorrhagic virus and the epidemic disease was rabbit plague.
