摘 要:讨论二冬膏对脂多糖诱发的大鼠急性肺损伤的保护作用。60只SD大鼠随机分为空白组、地塞米松组、模型组和二冬膏低、中、高剂量组,每组10只。地塞米松组给予5 mg/kg地塞米松灌胃,二冬膏低、中、高剂量组分别给予3 g/kg、6 g/kg和12 g/kg二冬膏灌胃。空白组和模型组每日灌胃等量生理盐水1次,连续11 d。除空白组外,其他各组通过鼻腔滴注脂多糖(5 mg/kg)造成急性肺损伤。使用HE染色法观察肺组织的病理变化,使用ELISA试剂盒测血清肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β)及肺组织丙二醛(MDA)和超氧化物歧化酶(SOD)浓度数值。将模型组与空白组比对,模型组大鼠血清中TNF-α、IL-6、IL-1β的浓度以及肺MDA浓度显著上调(P<0.01),而SOD的浓度显著下调(P<0.01);将各剂量组与模型组相比,二冬膏中、高剂量组大鼠血清中TNF-α浓度,二冬膏低、中、高剂量组大鼠血清中IL-6、IL-1β的浓度及肺组织MDA浓度显著下调(P<0.01),而SOD的浓度显著上调(P<0.01)。光镜下二冬膏各组肺组织水肿减轻,病变肺组织浸润情况改善。二冬膏可以通过减少炎症因子的释放、减轻氧化应激反应等作用,缓解肺损伤,对LPS诱导的急性肺损伤起到一定的保护作用。The purpose of this dissertation is to investigate Erdong ointment's proctive effect of rat's lung during the lipopolysaccharide-induced acute injur.Sixty SD rats were randomly divided into blank group,dexamethasone group,model group and Erdong ointment low,medium and high dose groups,with 10 rats in each group.The dexamethasone group's rats were given intragastric administration of 5 mg/kg dexamethasone,and the low,medium and high dose groups of Erdonggao's rats were given intragastric administration of 3 g/kg,6 g/kg and 12 g/kg Erdonggao respectively.The blank group's rats and model group's rats were given the same amount of normal saline once a day during 11 days.Except for the blank group,acute lung injury was induced by intranasal instillation of lipopolysaccharide(5 mg/kg).HE staining was used to observe the pathological changes of lung tissue of rats,and ELISA kit was used to measure the concentrations of serum tumor necrosis factor-α( TNF-α),interleukin-6(IL-6),interleukin-1β( IL-1β),malondialdehyde(MDA) and superoxide dismutase(SOD) in lung tissue.Compared with the blank group,the concentrations of TNF-α,IL-6,IL-1β in serum and MDA in lung of the model group were significantly increased(P<0.01),while the concentration of SOD was significantly decreased(P<0.01).Compared with the model group,the concentration of TNF-α in serum of rats in middle and high dose groups of Erdong ointment,the concentration of IL-6,IL-1β in serum and MDA in lung tissue of rats in low,middle and high dose groups of Erdong ointment were significantly decreased(P<0.01),while the concentration of SOD was significantly increased(P<0.01).Under light microscope,the edema of lung tissue was alleviated and the infiltration of lung tissue was improved.Erdong ointment can protect lung by reducing the release of inflammatory factors and reducing oxidative stress response,and play a protective role in LPS-induced acute lung injury.
