摘  要:目的观察应激性高血压(SIH)大鼠延髓胱硫醚-β-合酶(CBS)和3-巯基丙酮酸转硫酶(3-MPST)/硫化氢(H_(2)S)合成途径的变化。方法18只雄性WKY大鼠按照随机数字表法分为对照组(n=6)和SIH组(n=12)。对照组大鼠不做任何处理,SIH组大鼠采用电击足底结合噪音刺激制作SIH模型。四道生理记录仪测定两组大鼠血压和心率,亚甲蓝法测量两组延髓H 2S生成,蛋白质免疫印迹法检测对照组和SIH组大鼠延髓CBS和3-MPST蛋白的变化。结果对照组大鼠的收缩压和心率分别是(119.21±6.49)mmHg和(326.40±13.54)次/min,SIH组大鼠的收缩压和心率分别为(147.16±9.76)mmHg、(402.51±19.28)次/min。与对照组比较,SIH组大鼠的收缩压和心率分别增加了23.45%和23.35%,差异均有统计学意义(P<0.05)。对照组和SIH组大鼠延髓H 2S生成率分别是(1492.76±121.67)pmol·min^(-1)·mg^(-1)、(247.96±107.17)pmol·min^(-1)·mg^(-1),与对照组比较,SIH组大鼠延髓H 2S生成率显著降低,差异有统计学意义(P<0.05)。对照组大鼠延髓中CBS和MPST蛋白表达水平分别为2.98±0.43、0.77±0.14。应激刺激后,SIH组大鼠延髓CBS和MPST蛋白表达水平分别为2.20±0.18、0.56±0.16。与对照组比较,SIH组大鼠延髓CBS蛋白表达水平降低,差异有统计学意义(P<0.05),MPST蛋白表达比较,差异无统计学意义(P>0.05)。结论SIH时H 2S生成减少,其机制可能是延髓H 2S生成酶CBS和3-MPST蛋白表达水平降低。Objective To assess cystathionine beta-synthase(CBS)and 3-mercaptopyruvate sulfurtransferase(3-MPST)in endogenous hydrogen sulfide(H_(2)S)biosynthesis in the medulla of stress-induced hypertension(SIH)rats.Methods Eighteen male WKY rats were randomly divided into control group(n=6)and SIH group(n=12)according to the random number table method.The control group rats did not receive any treatment,while the SIH group rats used electric shock to the soles of their feet combined with noise stimulation to create an SIH model.The blood pressure and heart rate was determined by four channel physiological recorder,we used a methylene blue(MB)method to quantify the generation rate of H 2S and Western blotting analysis was conducted to measure the changes of CBS and 3-MPST in the medulla in both groups.Results The systolic blood pressure and heart rate of the control group were(119.21±6.49)mmHg and(326.40±13.54)beats/min,respectively,the systolic blood pressure and heart rate of the SIH group rats were(147.16±9.76)mmHg and(402.51±19.28)beats/min,respectively.Compared to the control group,SBP and HR of the SIH group were increased by 23.45%and 23.35%,respectively,the differences were statistically significant(P<0.05).The generation rate of H 2S was(1492.76±121.67)pmol·min^(-1)·mg^(-1)in the control group and(247.96±107.17)pmol·min^(-1)·mg^(-1)in the SIH group,the difference was statistically significant(P<0.05).The expression levels of CBS and MPST proteins in the medulla oblongata of control group rats were 2.98±0.43 and 0.77±0.14,respectively.After stress stimulation,the expression levels of CBS and MPST proteins in the medulla oblongata of SIH group rats were 2.20±0.18 and 0.56±0.16,respectively.Compared with the control group,the expression level of CBS protein in the medulla oblongata of SIH group rats decreased,the difference was statistically significant(P<0.05),while the expression of MPST protein showed no statistically significance(P>0.05).Conclusion The study shows that central H 2S generation declin
