摘 要:目的 构建壳聚糖(CS)/骨形态发生蛋白2(BMP2)质粒温敏水凝胶复合体CS/壳聚糖纳米粒(CSn)(pDNA-BMP2)-GP,研究其生物相容性及其在大鼠颅骨缺损再生中的作用。方法 将1 mL CS/CSn-GP复合体注入18只SD大鼠左、右后肢大腿肌袋内,于第3天、第1、2、4、6、9周随机处死3只大鼠,取注射部位及邻近组织,苏木精和伊红染色后观察其炎症反应。将32只SD大鼠按照随机数字表法分为两组:CS/CSn(pDNA-BMP2)-GP组、CS/CSn-GP组,各16只。在大鼠颅骨人字缝的左右两侧,各做一直径为5 mm的骨缺损,在每只实验鼠左侧的颅骨缺损区不放入任何材料,作为空白组,右侧分别植入各组材料。术后分别于第4、8周时随机处死6只大鼠,取出样本进行CT测量颅骨缺损面积及苏木精和伊红染色评价骨再生情况。结果 苏木精和伊红切片显示,复合体注入肌袋后,未见明显炎症反应;术后第4、8周,在骨修复质量及速度上,CS/CSn(pDNA-BMP2)-GP组明显优于空白组和CS/CSn-GP组。术后第4、8周,颅骨缺损面积CT结果显示:CS/CSn(pDNA-BMP2)-GP组骨缺损面积分别为(0.06683±0.00248)、(0.02017±0.00816)S/cm^(2),均明显低于空白组[(0.15033±0.01748)、(0.05150±0.01183)S/cm^(2)]和CS/CSn-GP组[(0.12650±0.00706)、(0.03883±0.00422)S/cm^(2)],差异均有统计学意义(P<0.05)。结论 CS/CSn-GP复合修复体系组织相容性良好,可作为良好的组织工程支架。壳聚糖/BMP-2质粒温敏水凝胶复合修复体系CS/CSn(pDNA-BMP2)-GP可以诱导大鼠颅骨临界尺寸骨缺损内的新骨形成,骨质量优于其他组。Objective To prepared the injectable chitosan(CS)/bone morphogenesis protein 2(BMP2)/chitosan nanoparticles(CSn)(pDNA-BMP2)-GP thermosensitive hydrogel scaffold system,and evaluate its regenerative effects on the endogenous repair of rat critical-sized calvarial defects.Methods Eighteen SD rats were injected with 1 mL of CS/CSn GP complex into the thigh muscle bags of the left and right hind limbs.Three rats were euthanized at the 3rd day,1st,2nd,4th,6th,and 9th week.The injection site and adjacent tissues were taken and observed for inflammation after haematoxylin and eosin staining.Thirty-two SD rats were divided into two groups by the random number table method:CS/CSn(pDNA-BMP2)GP group and CS/CSn GP group,with 16 rats in each group.A 5 mm diameter bone defect was made on both sides of the rat skull.No material was put into the left side of the skull defect area of each xperimental rat,which was used as a blank control group.Materials were implanted into the right side of each group,which was used as treatment group.Six rats were killed at random at 4 and 8 weeks after operation,and the samples were taken for CT measurement of the skull defect area and Haematoxylin and eosin staining to evaluate the bone regeneration.Results Haematoxylin and eosin sections showed that there was no obvious inflammatory reaction after the complex was injected into the muscle bag;at the 4th and 8th weeks after operation,the CS/CSn(pDNA-BMP2)GP group was significantly better than the blank group and CS/CSn-GP group in terms of bone repair quality and speed.At the 4th and 8th weeks after operation.At the 4th and 8th weeks after surgery,the CT results of the skull defect area showed that the bone defect area in the CS/CSn(pDNA-BMP2)-GP group was(0.06683±0.00248),(0.02017±0.00816)S/cm^(2),respectively,which were lower than those in the blank group[(0.15033±0.01748),(0.05150±0.01183)S/cm^(2)]and CS/CSn-GP group[(0.12650±0.00706),(0.03883±0.00422)S/cm^(2)],the differences were statistically significant(P<0.05).Conclusion CS/CSn G
