摘  要:【目的】为研究NtGCN2过表达对烟草耐盐性的影响,探究其参与盐胁迫应答机理。【方法】采用不同浓度Na Cl溶液处理NtGCN2转基因烟草植株,测定其生理生化指标和耐盐相关基因的表达,探究NtGCN2参与调控盐胁迫的生理与分子机制。【结果】(1)NtGCN2基因表达受到盐胁迫(100 mmol·L^(-1)Na Cl)的调控,在烟草中过表达NtGCN2可以提高转基因烟草的耐盐性。(2)盐胁迫下NtGCN2的过表达能够维持转基因烟草植株较高的光合作用速率和叶绿素含量,提高抗氧化相关基因表达和酶活性,增加了游离脯氨酸含量,降低了MDA的含量。(3)NtGCN2通过提高耐盐相关基因的表达和调节Na^(+)/K^(+)离子平衡增强转基因植株的耐盐性。【结论】NtGCN2通过提高烟草抗氧化能力、维持较高的光合作用速率和叶绿素含量、增强耐盐相关基因表达、调节Na^(+)/K^(+)平衡,改变植物生理状态,提高了转基因烟草的耐盐能力。[Objective]The present study aims to explore the effects of over-expression of NtGCN2 on salt tolerance as well as the possible mechanism of NtGCN2-mediated salt stress response.[Methods]NtGCN2 transgenic tobacco plants were treated with different concentrations of NaCl,the physiological and biochemical indexes were determined,and the salt tolerance-related genes were analyzed by qRT-PCR.[Results]Our results showed that NtGCN2 was regulated by 100 mmol・L^(-1)NaCl,and over-expression of NtGCN2 enhanced salt tolerance in tobacco.NtGCN2 could maintain higher photosynthetic rate and chlorophyll content,improve the gene expression of antioxidant-related genes and enzymatic activities,increase the free proline content,and decrease the content of MDA so as to enhance the salt-tolerance ability of transgenic tobacco plants under salt stress.Also,NtGCN2 could enhance salt-tolerance genes and maintain the steady state of Na^(+)/K^(+).[Conclusion]NtGCN2 enhances the salt tolerance of transgenic tobacco plants by enhancing leaf antioxidant ability,maintaining higher photosynthetic rate and chlorophyll content,facilitating salt tolerance-related genes expression,preserving cellular Na^(+)/K^(+)homeostasis,and changing the physiological status of tobacco.
