摘  要:【目的】为了探究烟草NtCycB2调控腺毛发生的可能分子机制。【方法】采用酶联免疫吸附法(ELISA)对K326、NtCycB2过表达株系NtCycB2-M和NtCycB2敲除株系NtCycB2-OE叶片内源激素进行测定;通过实时定量PCR分析NtCycB2对茉莉酮酸(JA)合成和信号途径相关基因表达量的影响,以及NtCycB2对外源激素茉莉酮酸甲酯(MeJA)的应答模式;NtCycB2-OE株系喷施不同浓度的MeJA,对其进行叶面腺毛形态观察和密度统计。【结果】(1)植物内源性激素检测结果表明敲除NtCycB2基因后烟叶JA含量显著增加,过表达NtCycB2基因后烟叶JA含量显著降低,其他内源激素的含量未发生显著变化。(2)NtCycB2基因影响JA合成、代谢和信号转导相关基因MFP、JMT和NtJAZ-1、-8、-15表达。(3)NtCycB2转录水平受外源激素MeJA的抑制。(4)MeJA促进NtCycB2-OE植株叶面长柄腺毛的发生。【结论】部分JA信号通路相关基因的表达受NtCycB2的影响,同时JA信号可以抑制NtCycB2的表达,进而促进长柄腺毛的发生。[Objective]This study aims to explore the possible molecular mechanism of NtCycB2 in regulating glandular trichomes generation in tobacco.[Methods]Enzyme linked immunosorbent assay(ELISA)was used to determine the endogenous hormones in leaves of K326,NtCycB2-M and NtCycB2-OE.The effects of NtCycB2 on the expression of genes related to JA signal pathways as well as the response pattern of NtCycB2 to exogenous hormone MeJA were analyzed by qPCR.NtCycB2-OE strain was sprayed with MeJA at different concentrations,and the morphology and density of leaf glandular trichomes were observed.[Results](1)Comparison of plant endogenous hormone contents in K326,NtCycB2-M and NtCycB2-OE showed that the JA content increased significantly in NtCycB2-M,while decreased significantly in NtCycB2-OE,and the contents of other endogenous hormones did not significantly change.(2)NtCycB2 gene affected the gene expressions related in JA synthesis,metabolism and signal transduction,such as MFP JMT and NtJAZ-1,-8,-15.(3)NtCycB2 transcription levels were inhibited by MeJA.(4)MeJA promoted the occurrence of long stalked glandular trichomes in NtCycB2-OE plants.[Conclusion]The expression patterns of some JA signaling genes were affected by NtCycB2,and JA signaling can inhibit NtCycB2 expression and promote the occurrence of long stalk glandular trichomes.
