籽莲响应腐败病菌侵染早期的转录组分析

摘  要:由尖孢镰刀菌莲专化型引起的腐败病是籽莲主要病害之一。为了探明籽莲对莲腐败病菌侵染的响应分子机制,以寸三莲为研究材料,采用转录组测序技术对籽莲根系进行转录组测序,鉴定籽莲响应腐败病菌侵染的关键基因。结果表明,莲腐败病菌接种后第5天,籽莲根系共发现402个基因显著差异表达,包括107个MYB、WRKY等转录因子显著上调表达。KEGG富集分析表明,大量差异基因显著富集于与肉桂酸、类黄酮合成相关的苯丙烷、类黄酮生物合成代谢通路,初步推测Nn7g37110、Nn7g37111和细胞色素P450等相关功能基因与籽莲响应莲腐败病菌侵染有关。本研究结果以期为籽莲抗腐败病分子育种提供理论参考。Rot disease caused by Fusarium oxyporum f.sp.nelumbicola is one of the main diseases of seed lotus.In order to elucidate the molecular mechanism behind the response to Fusarium oxyporum f.sp.nelumbicola,taking Cun Sanlian as the research material,the transcriptome analysis of Cun Sanlian roots under Fusarium oxyporum f.sp.nelumbicola infection was performed to identify the key genes of seed lotus responding to the infection of rot pathogen.The results showed that on the 5th day after inoculation,402 genes were significantly differentially expressed in seed lotus roots,including 107 transcription factors such as MYB,WRKY and so on.KEGG enrichment analysis showed that a large number of differential genes were significantly enriched in phenylpropane and flavonoid biosynthesis metabolic pathways related to cinnamic acid and flavonoid synthesis.It was speculated that Nn7g37110,Nn7g37111 and cytochrome P450 genes were related to lotus infection.This study could provide theoretical references for molecular breeding of resistance to corruption in seed lotus.
