菟丝子总黄酮三阴交穴位贴敷对流产大鼠妊娠结局及Th1/Th2细胞因子平衡的影响

摘　　要：目的:探究菟丝子总黄酮三阴交贴敷对流产大鼠妊娠结局及Th1/Th2细胞因子平衡的影响。方法:将确认受孕的SPF级SD雌鼠随机分为正常组、模型组、地屈孕酮组、菟丝子总黄酮组、菟丝子总黄酮三阴交贴敷组,每组15只。妊娠第1天开始,地屈孕酮组灌胃地屈孕酮水溶液5 mg/kg并于三阴交处贴敷淀粉膏贴,菟丝子总黄酮组灌胃菟丝子总黄酮200 mg/kg并于三阴交处贴敷淀粉膏贴,菟丝子总黄酮三阴交贴敷组灌胃等体积蒸馏水并于三阴交处贴敷菟丝子总黄酮膏贴,正常组、模型组灌胃等体积蒸馏水并于三阴交处贴敷淀粉膏贴,每日1次,持续11天。除正常组外,其余4组孕鼠于孕11天时灌服米非司酮4 mg/kg诱导建立SD大鼠的流产模型。实验结束后麻醉大鼠采集眼静脉血,分离血清检测催乳素(PRL)、孕酮(P)含量;分离子宫蜕膜组织,免疫组化法检测蜕膜组织中孕激素受体(PR)的表达,ELISA法检测蜕膜组织中肿瘤坏死因子α(TNF-α)、γ干扰素(IFN-γ)、白细胞介素(IL)-4、IL-10含量,Western Blot法检测蜕膜组织TNF-α、IFN-γ、IL-4、IL-10及胎盘组织中转录因子T-bet、GATA结合蛋白3(GATA-3)蛋白表达。结果:与正常组比较,模型组大鼠血清中PRL、P含量、蜕膜组织中PR的表达、IL-4、IL-10含量与蛋白表达及胎盘组织中GATA-3表达明显降低(P<0.05),流产率、蜕膜组织中TNF-α、IFN-γ含量与蛋白表达及胎盘组织中T-bet表达明显升高(P<0.05);与模型组比较,地屈孕酮组、菟丝子总黄酮组、菟丝子总黄酮三阴交贴敷组大鼠血清中PRL、P含量、蜕膜组织中PR的表达、IL-4、IL-10含量与蛋白表达及胎盘组织中GATA-3表达明显升高(P<0.05),流产率、蜕膜组织中TNF-α、IFN-γ含量与蛋白表达及胎盘组织中T-bet表达明显降低(P<0.05),且菟丝子总黄酮三阴交贴敷组和地屈孕酮组各项指标较菟丝子总黄酮组变化更明显(P<0.05)。结论:菟丝子总黄酮三阴交Objective:To investigate the effect of dodder total flavonoids and Sanyinjiao acupoint application on pregnancy outcomes and Th1/Th2 cytokine balance in aborted rats.Methods:The pregnant SPF SD female rats were randomly divided into normal group,model group,dydrogesterone group,dodder total flavonoid group and dodder total flavonoid Sanyinjiao acupoint application group,with 15 rats in each group.From the first day of pregnancy,dydroprogesterone group was administered with 5 mg/kg of dydroprogesterone aqueous solution by gavage and applied with starch paste at Sanyinjiao.Dodder total flavonoid group was administered with 200 mg/kg of dodder total flavonoid by gavage and applied with starch paste at Sanyinjiao.Dodder total flavonoid and Sanyinjiao acupoint application group was administered with equal volume of distilled water by gavage and applied with Cuscuta total flavonoid paste at Sanyinjiao.Normal group and model group were administered with equal volume of distilled water by gavage and applied with starch paste at Sanyinjiao once a day for 11 days.Except for the normal group,the other 4 groups of pregnant rats were given mifepristone 4 mg/kg by gavage on the 11th day of pregnancy to induce the establishment of an abortion model in SD rats.After the experiment,the ocular venous blood was collected from anesthetized rats,and the serum was separated for the detection of prolactin(PRL)and progesterone(P);the decidual tissue of the uterus was separated,the expression of progesterone receptor(PR)in decidua tissue was measured by immunohistochemistry,the tumor necrosis factorα(TNF-α),interferonγ(IFN-γ),interleukin(IL)-4 and IL-10 in decidua tissue were measured by ELISA;the protein expression of TNF-α,IFN-γ,IL-4 and IL-10 in decidual tissue and transcription factors T-bet,GATA-binding protein 3(GATA-3)in placental tissue was measured by Western Blot.Results:Compared with the normal group,the levels of PRL and P in serum,the expression of PR in decidua tissue,the content of IL-4 and IL-10 and protein expressi
