摘 要:在单因素实验的基础上,以五味子乙素得率为响应值,采用响应面法优化了酶解-超声双辅助双水相体系制备五味子提取物的工艺条件,并比较了双水相体系、酶辅助双水相体系、超声辅助双水相体系及酶解-超声双辅助双水相体系等方法对五味子乙素的提取效果。结果表明,酶解-超声双辅助双水相体系制备五味子提取物的最佳条件为:乙醇体积分数27.45%、硫酸铵质量分数25.82%、超声时间56 min、液固比30.98∶1(mL∶g),在此条件下,五味子乙素得率为1.77 mg·g^(-1),与模型预测值(1.76 mg·g^(-1))接近;酶解-超声双辅助双水相体系制备的五味子乙素得率显著高于双水相体系、酶辅助双水相体系及超声辅助双水相体系,说明前处理可有效促进双水相体系对五味子乙素的富集。Taking the yield of schisandrin B as a response value,we optimized the preparation process of Schisandra chinensis(Turcz.)Baill.extract by response surface methodologies on the basis of single-factor experiments.Moreover,we compared the extraction efficiency of schisandrin B by aqueous two-phase system,enzyme-assisted aqueous two-phase system,ultrasonic-assisted aqueous two-phase system,and enzymolysis-ultrasonic-assisted aqueous two-phase system.The results show that the optimal process of enzymolysis-ultrasonic-assisted aqueous two-phase system is determined as follows:the ethanol volume fraction of 27.45%,the ammonium sulfate mass fraction of 25.82%,the ultrasonic time of 56 min,and the liquid-solid ratio of 30.98∶1(mL∶g).Under above conditions,the yield of schisandrin B can reach 1.77 mg·g^(-1),which is close to the predicted value(1.76 mg·g^(-1)).The yield of schisandrin B by enzymolysis-ultrasonic-assisted aqueous two-phase system is significantly higher than that by aqueous two-phase system,enzyme-assisted aqueous two-phase system,and ultrasonic-assisted aqueous two-phase system,indicating that pretreatment can effectively promote the enrichment of schisandrin B by the aqueous two-phase system.
