摘 要:【目的】探明黄岩凤仙花(Impatiens huangyanensis)atpB-rbcL序列的特征及其分类地位,为后续开展遗传结构和遗传多样性研究奠定基础。【方法】在野外采样后利用PCR方法克隆黄岩凤仙花的atpB-rbcL全长,借助生物信息学手段进行序列分析,并构建系统发育树。【结果】黄岩凤仙花atpB-rbcL序列的全长为764 bp,GC值为27.5%;与淡黄绿凤仙花(I.chloroxantha)、阔萼凤仙花(I.platysepala)和武夷凤仙花(I.wuyiensis)的相似性最高,达98%以上;经多重比对,凤仙花属植物atpB-rbcL存在较多的插入、缺失、转换和颠换现象;经系统发育分析,黄岩凤仙花与淡黄绿凤仙花、阔萼凤仙花和武夷凤仙花处于同一组,亲缘关系最接近。【结论】叶绿体atpB-rbcL具有较高的变异,可用于黄岩凤仙花的分子鉴定。【Objective】The characteristics and taxonnomic status of atpB-rbcL sequence from Impatiens huangyanensis were determined,which laid foundation for the following research of genetic structure and diversity.【Method】Based on field sampling,the full length of atpB-rbcL was isolated by PCR,and sequence analysis was performed by bioinformatics tools,and then a phylogenic tree was constructed.【Result】The full atpB-rbcL was 764 bp in length,with a GC content of 27.5%.The sequence shared a high similarity of more than 98%with I.chloroxantha,I.platysepala,and I.wuyiensis.Multiple sequence alignment results revealed a number of insertions,deletions,transversions,and transitions in atpB-rbcL.Phylogenic analysis showed that I.huangyanensis,I.chloroxantha,I.platysepala,and I.wuyiensis shared the same clade,revealing their closest relationships.【Conclusion】High variations were observed in the chloroplast atpB-rbcL sequences,which could be used for molecular identification of I.huangyanensis.
