白藜芦醇对镧神经损伤仔鼠脑内HIF-1α表达的影响

摘  要:为探讨白藜芦醇对镧神经损伤仔鼠的作用及其对脑内低氧诱导因子1-α(HIF-1α)表达的影响,建立仔鼠5 g/L LaCl_(3)水溶液镧暴露模型。建模后把仔鼠随机分为对照组、阳性对照组和低、中、高剂量白藜芦醇干预组,分别以蒸馏水或不同剂量的白藜芦醇灌胃4 w,并在次日检测各组仔鼠学习及记忆能力。白藜芦醇干预前后称量仔鼠体质量、全脑质量、海马质量,计算全脑系数、海马系数并测定海马组织HIF-1αmRNA和蛋白含量。结果显示:染镧仔鼠的体质量下降、脑发育受损;白藜芦醇干预后染镧仔鼠的体质量、全脑质量、海马质量及其组织系数均逐渐上升;随着白藜芦醇干预剂量的增加,定向航行实验中仔鼠的游泳距离和平均逃避潜伏期逐渐缩短(P<0.05),空间探索实验中仔鼠游泳距离及停留在目标象限的时间、穿越目标象限和隐蔽站台的次数均逐渐增加(P<0.05);海马组织中HIF-1αmRNA表达水平明显上升(P<0.05),HIF-1α蛋白的含量明显增加(P<0.05)。因此,镧可在仔鼠脑组织蓄积并造成其学习、记忆能力下降,白藜芦醇可减轻镧暴露所致神经损伤,其机制可能与海马组织中HIF-1α基因含量的增加有关。In order to investigate the effect of resveratrol on lanthanum-induced nerve injury and the expression of hypoxia inducible factor 1-α(HIF-1α)in the brain of offspring rats,a model of lanthanum exposure in 5 g/L LaCl_(3)aqueous solution was established.After modeling,the offspring rats were randomly divided into control group,positive control group as well as low,medium and high dose resveratrol intervention groups.They were given distilled water or different doses of resveratrol for 4 weeks,and the learning and memory ability of offspring rats in each group was detected the next day.The body mass,whole brain mass and hippocampus mass of offspring rats were measured before and after resveratrol intervention.The whole brain coefficient and hippocampus coefficient were calculated,and the HIF-1αmRNA and protein contents in hippocampus were determined.The results showed that the body mass of lanthanum-exposed offspring decreased and the brain development was impaired.After resveratrol intervention,the body mass,whole brain mass,hippocampus mass and tissue coefficient of lanthanum-exposed offspring rats increased gradually.With the increase of resveratrol intervention dose,the swimming distance and average escape latency of the offspring rats in the directional navigation experiment were gradually shortened(P<0.05),and the swimming distance and the time of staying in the target quadrant,the number of crossing the target quadrant and the hidden platform in the space exploration experiment were gradually increased(P<0.05).The expression level of HIF-1αmRNA in hippocampus was significantly increased(P<0.05),and the content of HIF-1αprotein was significantly increased(P<0.05).Therefore,lanthanum can accumulate in the brain tissue of offspring rats,resulting in decreased learning and memory ability.Resveratrol can alleviate nerve injury caused by lanthanum exposure.The mechanism may be related to the increase of HIF-1αgene content in hippocampus.
