摘 要:目的探索纳米银薇乔线用于小鼠背部皮内连续缝合线道周围炎性细胞和炎性蛋白的表达情况,分析其近期抗炎疗效。方法对4-0普通薇乔线采用化学涂层技术制备纳米银薇乔线。18只10~12周Balb/C小鼠随机分为3组(每组各6只),剃除小鼠背部毛发,纵行切开15 mm皮肤全层切口。分别采用4-0带针普通薇乔线、抗生素薇乔线、纳米银薇乔线行皮内连续缝合。术后5 d,以纵形切口为中心,取材全层皮肤组织。采用免疫组织化学染色方法比较线道周围中性粒细胞、巨噬细胞、白细胞介素-6(interleukin-6,IL-6)、肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)的表达水平,并计算平均光密度(mean optical density,MOD)。原位末端转移酶标记(mediated dUTP-biotin nick end labeling,TUNEL)染色了解线道周围细胞凋亡情况。Masson染色了解线道周围胶原蛋白沉积情况。统计学方法采用单因素方差分析和student’s t检验。结果免疫组织化学染色显示,普通薇乔线、抗生素薇乔线和纳米银薇乔线的中性粒细胞MOD值为0.523±0.137、0.412±0.122、0.229±0.079;巨噬细胞MOD值为1.125±0.126、0.585±0.142、0.209±0.078;IL-6的MOD值为1.137±0.168、0.652±0.131、0.227±0.086;TNF-α的MOD值为1.219±0.124、0.573±0.109、0.179±0.045,纳米银薇乔线线道周围炎性细胞浸润、IL-6和TNF-α的表达最轻微,普通薇乔线表达最明显,3组间比较差异均有统计学意义(P值均<0.001)。TUNEL染色显示,普通薇乔线、抗生素薇乔线和纳米银薇乔线凋亡细胞的MOD值为0.167±0.035、0.478±0.151、0.125±0.035,纳米银薇乔线线道周围凋亡细胞最少,抗生素薇乔线凋亡细胞最多,3组间比较差异均有统计学意义(P值均<0.001)。Masson染色显示,纳米银薇乔线线道周围胶原蛋白沉积显著,抗生素薇乔线线道周围未见胶原蛋白沉积。结论在小鼠皮内连续缝合模型上,纳米银薇乔线的近期抗炎疗效优于抗生素薇乔线Objective To investigate the expression of inflammatory cells infiltration and inflammatory proteins around silver nanoparticles coated Vicryl suture in mice dorsal intradermal running suture models,and to analyze the anti-inflammatory effect.Method The 4-0 Vicryl suture were coated with silver nanoparticles by chemical coating technique.Eighteen Balb/C mice at 10-12 weeks were randomly divided into 3 groups(with 6 mice in each group).The back hair of the mice was shaved and a 15 mm longitudinal full-layer incision was made.Continuous intradermal suture was performed using 4-0 common Vicryl suture,antibiotic Vicryl suture,and silver nanoparticles coated Vicryl suture,respectively.The whole skin tissue was collected with longitudinal incision as the center at 5 days after surgery.Immunohistochemical staining was used to compare the expression levels of neutrophils,macrophages,interleukin-6(IL-6),and tumor necrosis factor-α(TNF-α)around the channels.The mean optical density(MOD)was calculated.Mediated dUTPbiotin nick end labeling(TUNEL)staining was used to investigate the apoptosis of cells around the channels.Masson staining was used to investigate the collagen deposition around the channels.One-way analysis of variance and student't t-test were used for statistical analysis.Result The immunohistochemistry staining indicated neutrophil MOD in common Vicryl suture group,antibiotic Vicryl suture group and silver nanoparticles coated Vicryl suture groups was 0.523±0.137,0.412±0.122,0.229±0.079,respectively;macrophage MOD in above three groups was 1.125±0.126,0.585±0.142,0.209±0.078,respectively;IL-6 MOD in above three groups was 1.137±0.168,0.652±0.131,0.227±0.086,respectively;TNF-αMOD in above three groups was 1.219±0.124,0.573±0.109,0.179±0.045,respectively;the infiltration of inflammatory cells,expression of IL-6 and TNF-αwas most obvious in common Vicryl suture group and least obvious in silver nanoparticles coated Vicryl suture group;the differences among the three groups were statistically signi
