摘 要:目的:观察丹参酮ⅡA磺酸钠(sodium tanshinoneⅡA sulfonate,STS)对转换生长因子-β1(TGF-β1)诱导的心房纤维化效应的影响。方法:培养新生乳鼠心房成纤维细胞。MTT比色法测定心房成纤维细胞增殖率,[3H]脯氨酸掺入测定心房成纤维细胞合成胶原速率。共聚焦荧光显微镜及Western blot检测Ⅰ/Ⅲ型胶原表达、Smad2磷酸化及核转移。结果:TGF-β1处理24 h后,心房成纤维细胞增殖明显增强,同时Ⅰ/Ⅲ型胶原表达及Smad2磷酸化/核转移亦明显增强。丹参酮ⅡA磺酸钠各浓度组以上效应均减弱。结论:丹参酮-ⅡIA磺酸钠能明显减轻TGF-β1诱导的心房纤维化效应,机制与调控Smad2磷酸化及核转移相关。OBJECTIVE To investigate the functions of sodium tanshinoneⅡA sulfonate(STS)on TGF-β1-induced the activation of TGF-β1 and syntheses collagen protein in atrial fibroblasts(AFs).METHODS In the primary culture of neonatal rat AFs,as indexes of myocardial fibrosis,the cell proliferation was determined by MTT.Collagen synthesis rate was measured by[3H]proline incorporation.The level ofⅠ/Ⅲcollagen protein was determined by Western blot.Smad2 phosphorylation/nuclear translocation was measured by fluorescent confocal microscopy and Western blot.RESULTS TGF-β1 could increase the level ofⅠ/Ⅲcollagen protein,cell proliferation.Furthermore,TGF-β1 could increase Smad2 phosphorylation/nuclear translocation.After treatment of STS,these indexes were obviously downregulated.CONCLUSION STS can decrease TGF-β1-induced atrial fibrosis in atrial fibroblasts,through inhibiting Smad2 phosphorylation/nuclear translocation.
