摘 要:大豆种子在长期储存条件下发生老化,导致种子的品质降低、萌发力下降、幼苗活力变差,最终造成大豆大规模减产。种子耐储藏性是一个复杂的数量性状,受多基因控制。为了探究大豆种子耐储藏性机理,挖掘相关基因,以中豆27和九农20及其杂交衍生的112份重组自交系(Recombinant Inbred Lines,RIL)群体为材料,评价大豆种子耐储藏性,利用343907个高质量的SNPs标记构建Binmap,定位大豆种子耐储藏相关性状的QTLs,并利用基因组、转录组、代谢组进一步分析。结果显示:本研究共得到31个QTLs,分布于大豆的10条染色体上。多组学联合分析共发现与种子耐储藏性相关的3个候选基因,并发现种子耐储藏性相关的通络包括黄酮类生物合成途径和亚油酸代谢等。研究结果为深入解析大豆种子耐储藏遗传本质和大豆耐储藏品种的培育提供理论依据和技术支持。The aging of soybean seeds under long-term storage conditions leads to the reduction of seed quality,germination ability and seedling vitality,and finally leads to the large-scale yield reduction of soybean.Seed storage-tolerance is a complex quantitative trait controlled by multiple genes.In order to explore the mechanism of soybean seed storage tolerance,the relevant genes were mined.In this study,112 recombinant inbred lines derived from the cross of Zhongdou 27 and Jiunong 20 were used as materials to evaluate the storage-tolerance of soybean seeds.Binmap was constructed using 343907 high-quality SNPs markers,QTLs for storage related traits of soybean seeds were mapped and further analyzed by genome,transcriptome and metabolome.A total of 31 QTLs were obtained in this experiment,distributed on 10 chromosomes of soybean.Through the combined analysis of genome,transcriptome and metabolome,3 candidate genes related to seed storage tolerance were found,covering 19 metabolic pathways including flavonoid biosynthesis pathways and linoleic acid metabolism etc.The results provided theoretical basis and technical support for further analyzing the genetic architecture of soybean seed storage tolerance and breeding soybean storage tolerance varieties.
