摘 要:以普通油茶(Camellia oleifera)优良无性系‘闽43’为实验材料,进行生理试验和酶活性检测,采用定量蛋白质组学技术对油茶成熟过程中3个时期的籽仁进行分析,探讨油茶籽油脂代谢的调控机制。研究发现,共有284个蛋白差异表达,依据功能分为17类,其中脂质代谢与运输相关蛋白多数呈现下调表达。硬脂酰-ACP去饱和酶(SAD)和胆碱激酶(CK)蛋白表达量的下调,与不饱和脂肪酸生成速率的减缓有关;在油茶果实成熟过程中的第二个阶段,脂肪酶(LPS)活性的下调造成游离脂肪酸的产生减少。由于长链脂酰辅酶A合成酶(LACS)蛋白表达量的下调,亚油酸和亚麻酸以游离脂肪酸的形式存在较长时间,促使脂氧合酶(LOX)的活性上升,导致该时期亚油酸和亚麻酸含量的下降。研究结果表明,油茶籽成熟过程中,不饱和脂肪酸含量的调控与油脂代谢相关蛋白表达模式和酶活性的变化密切相关。研究结果为选育高品质的油茶品种提供理论依据。In order to explore the regulation mechanism of oil metabolism of Camellia oleifera,a superior clone of common camellia'Min 43'was used as experimental material to analyze the seed kernel of three stages during its maturation period by using quantitative proteomics technology through physiological test and enzyme activity detection.The results show that a total of 284 proteins were differentially expressed and divided into 17 categories according to their functions.Most of the proteins related to lipid metabolism and transportation were down-regulated.The down-regulation of the expression levels of stearyl-ACP desaturase(SAD)and choline kinase(CK)was related to the decrease of the rate of unsaturated fatty acid production.The down-regulation of lipase(LPS)activity during P2 reduced the production of free fatty acids.Due to the down-regulation of long-chain fatty acyl-CoA synthase(LACS)protein expression,both linoleic acid and linolenic acid would exist in the form of free fatty acids for a long time,which led to the increase of lipid oxygenase(Lox)activity and the decrease of linoleic acid content in this period.The results show that the regulation of unsaturated fatty acid content was closely related to the changes of lipid metabolization related protein expression patterns and enzyme activities during the maturation of C.oleifera seed.It provides theoretical basis for breeding high quality C.oleifera varieties.
