摘 要:目的 建立蚓激酶干粉水分、蛋白质和效价的快速测定方法。方法 将蚓激酶效价的琼脂糖-纤维蛋白平板法、测定水分的烘干法和测定蛋白质的凯氏定氮法测得的实验数据与近红外光谱结合化学计量学方法,建立快速定量分析模型,使用Micro NIR 1700光谱仪收集908~1 650 nm范围内的蚓激酶干粉光谱,每个光谱的13.6 ms积分时间对应于100次扫描的累积。结果 偏最小二乘回归(PLS)定量模型结果为,干燥失重中含水量的校正决定系数(R_(c)^(2))、预测决定系数(R_(p)^(2))、校正均方根误差(RMSEC)和预测均方根误差(RMSEP)分别为0.924 0、0.917 3、0.121 3%、0.127 5%;蛋白质含量的R_(c)^(2)、R_(p)^(2)、RMSEC和RMSEP分别为0.961 5、0.955 3、0.002 0、0.002 2;效价的R_(c)^(2)、R_(p)^(2)、RMSEC和RMSEP分别为0.963 2、0.959 1、26.971 3 IU/mg、28.587 6 IU/mg。结论 本方法操作简单,准确度高,结果可信,近红外分析技术是一种快速测定蚓激酶干粉中效价、水分及蛋白质的潜在工具。Objective To establish a rapid method for determination of moisture,protein and titer of lumbrokinase dry powder.Methods The experimental data obtained by the agarose fibrin plate method for lumbrokinase titer,the drying method for moisture determination and the Kjeldahl nitrogen determination method for protein determination were combined with the chemometrics method to establish a rapid quantitative analysis model.The spectrum of lumbrokinase dry powder in the range of 908~1 650 nm was collected by the Micro NIR 1700 spectrometer.The 13.6 ms integration time for each spectrum corresponds to the accumulation of 100 scans.Results Partial least squares regression (PLS) quantitative model results show that the water content R_c^(2),R_p^(2),RMSEC and RMSEP in dry weight loss are 0.924 0,0.917 3,0.121 3%and 0.127 5%,respectively.The R_c^(2),R_p^(2),RMSEC and RMSEP of protein content were 0.961 5,0.955 3,0.002 0 and 0.002 2,respectively.The titers of R_c^(2),R_p^(2),RMSEC and RMSEP were 0.963 2,0.959 1,26.971 3 IU/mg and 28.587 6 IU/mg,respectively.Conclusion The method is simple,accurate and reliable.Near infrared analysis is a potential tool for rapid determination of titer,moisture and protein in lumbrokinase dry powder.
