天麻钩藤饮对高血压前期大鼠血管内皮功能及miRNA表达影响的研究

摘　　要：目的:观察天麻钩藤饮干预高血压前期SHR大鼠对血压、血管内皮功能及相关miRNA的影响。方法:4周SHR大鼠40只分为模型组及中药高中低剂量组各10只,并予以同龄WKY大鼠10只进行对照,模型组和对照组不进行药物干预,每日灌胃与实验组相同体积的生理盐水;中药组分别予以不同剂量天麻钩藤饮灌胃,1个疗程8周,检测大鼠鼠尾血压、ELISA法检测血管内皮炎性因子TNF-α、VEGF,Western blot检测大鼠肾脏组织AT1受体及AngII,RT-PCR检测大鼠肾脏miR155-5P及miR122-5P的表达。结果:给药8周后给药组大鼠与模型组大鼠比较,血压升高幅度更小。各中药给药组大鼠内皮功能障碍相关指标TNF-α和VEGF降低,RAAS系统AT1和AngII的表达下降。SHR大鼠肾脏组织miRNA122-5p和miRNA155-5p的表达升高,而中药天麻钩藤饮的干预后miRNA122-5p表达降低,miRNA155-5p的表达有降低趋势。结论:在高血压前期予以天麻钩藤饮,可以有效减低高血压进程中血管内皮损伤,可能有延缓高血压进展、保护靶器官等作用。Objective:To observe the effect of Tianma Gouteng yin on blood pressure,vascular endothelial function and related miRNA of SHR rats intervened by prehypertension.Methods:SHR rats of four weeks old were divided into model group and TCM high-,medium-and low-dose group,and controlled with WKY of the same age.TCM groups were administered different doses of Tianma Gouteng yin respectively by gavage,the course of treatment was 8 weeks.The blood pressure of rat's tail was detected.Vascular endothelia inflammatory factors TNF-αand VEGF were detected by ELISA.AT1 receptor and AngII in rats'kidney tissue were detected by Western blot.The expressions of miR155-5P and miR122-5P in rats'kidney were detected by RT-PCR.Results:After 8 weeks of administration,the blood pressure of rats in administration group increased less than that in model group.The related indexes such as TNF-αand VEGF of rats'endothelial dysfunction in each TCM administration group decreased.The expressions of AT1 and AngII in RAAS system decreased.The expressions of miRNA122-5p and miRNA155-5p in renal tissue of SHR rats increased,while the expression of miRNA122-5p decreased after the intervention by Tianma Gouteng yin of TCM,and the expression of miRNA155-5p showed a decreasing trend.Conclusion:To administer Tianma Gouteng yin during prehypertension can effectively reduce vascular endothelial injury in the process of hypertension,may delay the progression of hypertension,and protect target organs,etc.
