摘 要:目的探讨健脾清化方对高脂饮食诱导的肥胖小鼠胸主动脉内皮的保护作用及可能机制。方法采用高脂饲料喂养C57BL/6小鼠12周诱导肥胖小鼠模型,将小鼠分为模型组、健脾清化方组和盐酸二甲双胍组,以正常喂养小鼠作为正常组,每组6只。给药6周后,HE染色观察小鼠胸主动脉内皮结构变化,Masson染色观察胸主动脉纤维化改变,透射电镜观察胸主动脉内皮细胞超微结构,免疫组化检测胸主动脉血小板内皮细胞黏附分子-1(PECAM-1/CD31)、NOD样受体热蛋白结构域相关蛋白3(NLRP3)表达。结果与正常组比较,模型组小鼠胸主动脉局部内皮细胞脱落,内膜层增厚,有泡沫样细胞聚集,局部管壁平滑肌和弹力纤维萎缩,平滑肌细胞排列紊乱,胶原纤维增加,CD31表达降低,NLRP3表达显著升高(P<0.01);与模型组比较,健脾清化方组小鼠胸主动脉内皮结构基本完整,内皮细胞脱落减少,内膜及中膜胶原纤维减少,CD31表达有升高趋势,NLRP3表达显著降低(P<0.01)。结论健脾清化方可能通过下调NLRP3表达改善肥胖小鼠受损的血管内皮形态和超微结构,改善炎症状态,CD31可能是健脾清化方保护血管内皮结构和通透性的重要靶点。Objective To investigate the protective effects and possible mechanism of Jianpi Qinghua Prescription on thoracic aortic endothelium in high-fat diet-induced obesity mice.Methods C57BL/6 mice were fed a high-fat diet for 12 weeks to induce obesity.The obesity mice were then randomly divided into model group,Jianpi Qinghua Prescription group and metformin group.The mice fed a normal diet were set as the normal group,with 6 mice in each group.After 6-weeks administration,HE staining was used to observe the pathological changes of thoracic aortic endothelium,Masson staining was used to observe the fibers changes of thoracic aortic endothelium,transmission electron microscopy was used to observe the ultrastructure of endothelial cells of thoracic aorta,the expressions of CD31 and NLRP3 were detected by immunohistochemistry.Results Compared with the normal group,the mice in the model group showed endothelial denudation,intimal thickening,and foam cells aggregation,the smooth muscle and elastic fiber of local tube wall atrophy,and the smooth muscle cells were in disorder,the collagen fibers increased,the expression of CD31 decreased and the expression of NLRP3 significantly increased(P<0.01).Compared with the model group,the structure of thoracic aortic endothelium in Jianpi Qinghua Prescription group was almost intact,the endothelial cells were less stripped,the collagen fibers decreased in intima and medica layer,the expression of CD31 showed an upward trend,and the expression of NLRP3 decreased significantly(P<0.01).Conclusion Jianpi Qinghua Prescription can improve impaired endothelial morphology and ultrastructure of obesity mice,ameliorate the inflammatory state.CD31 protein may be an important target of Jianpi Qinghua Prescription to protect vascular endothelial structure and permeability.
