摘 要:目的探究电针心包经穴对大鼠脑缺血后神经修复的影响及作用机制。方法150只SD大鼠随机分为正常组、假手术组、模型组、心包经组和肺经组,以颈外动脉插入线栓法制备大脑中动脉栓塞(MCAO)大鼠模型。心包经组选取“天泉”“曲泽”“内关”“大陵”,肺经组选取“天府”“尺泽”“列缺”“太渊”,每日电针1次,干预6 d休息1 d,分别于3、7、14、21、28 d进行取材。HE染色观察大鼠缺血侧脑组织病理改变,免疫组化染色和Western blot检测脑组织髓鞘相关生长抑制因子A(Nogo-A)、Nogo受体1(NgR1)蛋白表达。结果与正常组、假手术组比较,模型组大鼠各时点缺血侧脑组织神经细胞消失或皱缩,细胞间距增大,Nogo-A和NgR1蛋白表达明显升高(P<0.01);与模型组比较,心包经组和肺经组大鼠缺血侧脑组织神经细胞数量增加,形态结构改善,各时点Nogo-A和NgR1蛋白表达明显降低(P<0.01,P<0.05);与肺经组比较,心包经组大鼠缺血侧脑组织Nogo-A和NgR1蛋白表达降低更明显(P<0.01,P<0.05)。结论电针心包经穴能在一定程度上调节MCAO模型大鼠神经轴突发芽和再生微环境,其机制与抑制不同时点缺血脑组织Nogo-A和NgR1蛋白表达有关。Objective To explore the effect and mechanism of nerve repair by electroacupuncture at acupoints of pericardium meridian after cerebral ischemia in rats.Methods Totally 150 SD rats were randomly divided into normal group,sham-operation group,model group,pericardial meridian group and lung meridian group.A rat model of middle cerebral artery occlusion(MCAO)was established by inserting a suture into the external carotid artery.The pericardium meridian group selected“Tianquan”(PC2),“Quze”(PC3),“Neiguan”(PC6)and“Daling”(PC7),and the lung meridian group selected“Tianfu”(LU3),“Chize”(LU5),“Lieque”(LU7)and“Taiyuan”(LU9).Electroacupuncture was performed once a day,6 days of intervention and 1 day of rest,materials were collected on 3,7,14,21,and 28 day of intervention,respectively.Pathological changes of ischemic brain tissue in rats were observed by HE staining,Nogo-A and NgR1 protein expressions in brain tissue were detected by immunohistochemical staining and Western blot.Results Compared with the normal group and the sham-operation group,the nerve cells in ischemic brain tissue of the rats in model group disappeared or shrunken at each time point,the distance between the cells increased,and the expressions of Nogo-A and NgR1 proteins significantly increased(P<0.01).Compared with the model group,the number of nerve cells in ischemic brain tissue of the rats in pericardium meridian group and lung meridian group increased,the morphological structure was improved,and the protein expressions of Nogo-A and NgR1 protein at each time point significantly decreased(P<0.01,P<0.05).Compared with the lung meridian group,the expressions of Nogo-A and NgR1 proteins in ischemic brain tissue of the pericardium meridian group significantly decreased(P<0.01,P<0.05).Conclusion Electroacupuncture stimulation at acupoints of pericardium meridian can regulate the neural axon sprouting and regeneration microenvironment in MCAO model rats,and its mechanism is related to the inhibition of Nogo-A and NgR1 protei
