摘  要:目的基于均匀设计法筛选益气温阳活血利水中药组分防治慢性心力衰竭(CHF)的最佳配比及配伍效应。方法采用主动脉弓缩窄法制备CHF大鼠模型,运用均匀设计法,以大鼠血清N端脑利钠肽前体(NT-proBNP)含量和射血分数(EF)为因变量,通过多元线性回归分析,筛选益气温阳活血利水中药组分最佳剂量配比,并对筛选结果进行实验验证,测定大鼠心重指数,HE及Masson染色观察心肌组织病理形态,ELISA检测血浆血管紧张素Ⅱ(AngⅡ)、醛固酮(ALD)及血清肌钙蛋白T(cTnT)含量。结果回归方程提示益气温阳活血利水中药组分最佳配比为:黄芪总皂苷60 mg/kg,丹参总酚酸10 mg/kg,葶苈子水提物190 mg/kg,桂皮醛100 mg/kg。验证实验结果显示,与模型组比较,给药组病理切片心肌细胞排列较整齐,胶原纤维堆积改善;心重指数明显减少(P<0.01);血浆AngⅡ、ALD及血清cTnT含量均明显升高(P<0.05,P<0.01)。结论应用均匀设计与药效分析法可优化益气温阳活血利水中药组分防治CHF的最佳剂量配比。Objective To explore the optimal dosage ratio and compatibility effect of TCM components of Yiqi Wenyang Huoxue Lishui(invigorating qi,warming yang,promoting blood circulation and inducing diuresis)for the prevention and treatment of chronic heart failure(CHF)based on uniform design method.Methods CHF rat model was established by aortic arch narrowing,and uniform design was used.The content of N-telencephalic natriuretic peptide precursor(NT-proBNP)and ejection fraction(EF)were used as dependent variables,through multiple linear regression analysis,the optimal proportion dosage of TCM components of Yiqi Wenyang Huoxue Lishui was selected,and the screening results were verified experimentally.The heart weight index of rats was measured,and the myocardial histomathological morphology was observed by HE and Masson staining.The contents of angiotensinⅡ(AngⅡ)and aldosterone(ALD)in plasma and troponin T(cTnT)in serum were determined by ELISA.Results The regression equation suggested that the best ratio of TCM components of Yiqi Wenyang Huoxue Lishui was 60 mg/kg Astragalus saponins,10 mg/kg total salvianolic acid,190 mg/kg aqueous extract from Descurainiae Semen Lepidii Semen,and 100 mg/kg cinnamaldehyde.The results showed that compared with the model group,the myocardial cells in pathological sections of the administration group were arranged more neatly,and the accumulation of collagen fibers was improved;heart weight index decreased significantly(P<0.01);the contents of AngⅡ,ALD in plasma and cTnT in serum significantly increased(P<0.05,P<0.01).Conclusion Uniform design and pharmacodynamic analysis can be used to optimize the optimal dosage ratio of TCM components of Yiqi Wenyang Huoxue Lishui for the prevention and treatment of chronic heart.
