摘  要:家蝇是公共卫生和畜牧业的重要害虫,抗药性已成为家蝇有效控制的重要障碍。以往的研究表明,家蝇羧酸酯酶MdαE7某些位点的突变可以导致对有机磷和拟除虫菊酯类杀虫剂的抗性,但我国家蝇种群MdαE7基因突变的类型、分布和不同等位基因的分子进化关系等相关信息还非常缺乏。本研究检测了我国5个省份家蝇样品的羧酸酯酶MdαE7基因突变的类型和分布状况,并对普遍存在的251位点突变的进化做了起源分析。结果表明,我国家蝇普遍存在MdαE7 G137D和W251L/S抗性突变,不同地区的抗性等位基因频率有所不同。除了常见的G137D和W251L/S外,在我国家蝇中还发现了4个其他氨基酸替换,即Y148F、P157S、S250T和E273D。基于55条MdαE7基因片段的DNA序列,鉴定出8种不同的单倍型。单倍型的分子进化分析表明MdαE7251位点的突变具有不同的进化起源。The house y(Musca domestica Linnaeus)is an important pest in public health and animal husbandry.Insecticide resistance is an obstacle to the control of house ies.Previous studies have demonstrated that some point mutations in carboxylesterase MdαE7 confer resistance to organophosphorus and pyrethroid insecticides in the house fly.However,knowledge of genetic mutations in the MdαE7 gene,their distribution and evolutionary origin in the field house fly populations in China is lacking.In this study,the occurrence of genetic mutations in MdαE7 in field house ies collected from five provinces across China were investigated.Three well recognized resistance-conferring mutations(G137D,W251L and W251S)were detected with varying frequencies in the five locations.Moreover,four additional amino acid replacements(Y148F,P157S,S250T and E273D)were observed in this study.From a total of 55 sequences of the MdαE7 gene fragments,eight haplotypes were identified.Phylogenetic analysis of haplotypes supported multiple origins of the resistance mutation at position 251 in MdαE7.
