摘 要:目的不同三硝基苯磺酸(2,4,6-trinitrobenzenesulfonic acid,TNBS)比例诱导Balb/c小鼠克罗恩病(Crohn disease,CD)模型的建立及评价。方法以完全随机区组化的方法将32只Balb/c小鼠分为4组,按水∶无水乙醇∶TNBS比值分为模型1组(2∶5∶3)、模型2组(1∶5∶4)及模型3组(0∶5∶5),每组8只。另8只健康小鼠作为对照组,以小鼠一般情况、体质量变化、结肠长度变化、粪便性状、结肠组织病理学评分等为指标评价不同比例TNBS诱导的小鼠CD模型进展并计算小鼠生存率。于建模后第7天取材进行结肠组织病理学检查及组织病理评分,取小鼠血清检测炎性细胞因子IL-6水平,同时测量小鼠结肠长度。结果模型3组小鼠生存率低,整个实验周期体质量持续下降,建模第3天即出现严重腹泻、便血等症状,7天后结肠长度显著缩短(P<0.001),组织损伤严重,组织病理学评分及IL-6水平均显著升高(P<0.001);模型1组小鼠第3天情况与模型3组相似,体质量在第3、4天显著下降(P<0.05)后有所回升,第7天镜下检查结肠组织损伤及炎症情况轻,结肠长度并无显著缩短;模型2组小鼠体质量、结肠长度及IL-6水平变化情况与模型3组相似,炎性细胞浸润及结肠局部损伤有大量炎性细胞浸润。结论模型2组(体积比1∶5∶4)小鼠生存率高,炎症轻重程度适中,病理变化情况稳定,是一种CD研究较好的模型制备方法。OBJECTIVE To establish and evaluate the Crohn disease(CD)model of Balb/c mice induced by different ratios of 2,4,6-trinitrobenzenesulfonic acid(TNBS).METHODS A total of 24 male Balb/c mice were divided into model group 1(2∶5∶3),model group 2(1∶5∶4)and model group 3(0∶5∶5)according to the water∶anhydrous ethanol∶TNBS ratio with 8 mice in each group.Another 8 healthy mice were selected as the control group,and the development of CD model induced by TNBS with different proportions was evaluated and the survival rate was calculated by using the general condition,body mass change,colon length change,fecal character,mortality and colon Histopathological score as indicators.On the 7th day after modeling,the samples were taken for colon histopathological examination and histopathological score.The levels of inflammatory cytokine IL-6 in mouse serum were detected and the length of colon was measured.RESULTS The survival rate of mice in model 3 groups was low,and the body weight of mice in model 3 groups continued to decline during the whole experiment period;on day 7 of modeling,colon length was significantly shortened(P<0.001),tissue damage was serious under microscope,histopathological scores and IL-6 levels were significantly increased(P<0.001).The body mass of mice in model group 1 was significantly decreased on day 3 and day 4(P<0.05)and then recovered.On day 7,colon tissue damage and inflammation were light,and colon length was not significantly shortened;the changes of body mass,colon length and IL-6 level of mice in model group 2 were similar to those in model group 3.Microscopic examination showed a large number of inflammatory cell infiltration and local damage of colon.CONCLUSION The mice in model 2 group(volume ratio 1∶5∶4)had high survival rate,moderate inflammation and stable pathological changes,which was a good model preparation method for CD research.
