摘 要:目的探究葛根中葛根素、大豆苷和大豆苷元3种异黄酮类成分对ox-LDL诱导的脂质沉积和LPS诱导的炎症的影响。方法采用MTT法确定药物浓度和作用时间。以ox-LDL诱导Raw264.7细胞建立泡沫细胞模型,油红O染色后观察细胞泡沫化情况,酶法检测TC和TG的含量。以LPS诱导Raw264.7细胞建立炎症模型,ELISA检测肿瘤坏死因子-α(TNF-α)和白介素1β(IL-1β)水平。结果经ox-LDL诱导后,模型组细胞内出现大量红色脂滴,细胞泡沫化明显,与模型组比较,3个化合物各浓度组的泡沫细胞中脂滴变小、颜色变浅。ox-LDL诱导后泡沫细胞中TC和TG水平显著升高,与模型组比较,3个化合物均可显著降低泡沫细胞中TC和TG水平。LPS诱导后模型组细胞内TNF-α和IL-1β大量分泌,与模型组比较,3个化合物均可显著抑制模型细胞TNF-α的分泌,葛根素和大豆苷元能显著抑制模型细胞IL-1β的分泌。结论葛根素、大豆苷和大豆苷元能明显抑制ox-LDL诱导的脂质沉积和LPS诱导的炎症因子分泌。OBJECTIVE To investigate the effects of puerarin,daidzin and daidzein from Puerarialobata(Willd.)Ohwi on lipid accumulation induced by oxidized low density(ox-LDL)and inflammation induced by lipopolysaccharide(LPS)in RAW264.7 cells.METHODS MTT assay was used to determine the drug concentration and action time.The RAW264.7 cells were induced by ox-LDL to establish foam cell.The concentration of lipid droplets in the foam cells was observed by oil red O staining,and,the levels of the total cholesterol(TC)and triglycerides(TG)were detected by enzyme method.The RAW264.7 cells were induced by LPS to release inflammatory factors,and the expressions of tumor necrosis factor(TNF-α)and interleukin 1β(IL-1β)were detected by the enzyme-linked immunosorbent assay(ELISA).RESULTS A large number of red lipid droplets appeared in the cells of the model group,after induced by ox-LDL,and the cell foaming was obvious.Compared with the model group,the lipid droplets in foam cells became smaller and lighter after treatment with the three isoflavones in different concentrations.After induced by ox-LDL,the TC and TG content in foam cells increased significantly.The three compounds significantly decreased the content of TC and TG in foam cells.The secretion of TNF-αand IL-1βwere increased in inflammatory cell that induced by LPS.The three isoflavones reduced the secretion of TNF-α,and puerarin and daidzein reduced the secretion of IL-1βin the inflammatory cells,significantly.CONCLUSION The three isoflavones could restrain the lipid deposition induced by ox-LDL,and reduce the secretion of inflammation factors induced by LPS.
