高效液相色谱法测定铁皮石斛中的5种核苷类物质含量

摘　　要：目的建立高效液相色谱法检测铁皮石斛中5种核苷类物质的分析方法。方法采用月旭C 18 Ultimate AQ-C 18(4.6×250 mm,5μm)色谱柱;流动相为20 mmol·L^(-1)乙酸铵水溶液(A)-甲醇(B),梯度洗脱;流速:1.0 mL·min^(-1),检测波长:260 nm,柱温:30℃,进样体积:20μL。结果5种核苷类物质在各自线性范围内线性良好,加样回收率为97.24%~100.10%之间,RSD小于2.12%。结论该方法操作简便、稳定可靠,可用于铁皮石斛中核苷类物质的测定。OBJECTIVE To establish a method for simultaneous determination of 5 nucleotides including cytosine,hypoxanthine,uridine,inosine and guanosine in the Dendrobium officinale by HPLC.METHODS The analysis was achieved with a Wech Ultimate AQ-C 18(4.6×250 mm,5μm)by gradient elution of 20 mmol·L^(-1) ammonium acetate aqueous solution(A)and acetonitrile(B).The flow rate was 1.0 mL·min^(-1) and the detection wavelength was 260nm.The column temperature was controlled at 30℃.RESULTS The curves of five target components showed a good linearity within their respective linear range.In addition,the recovery rates were 97.24%-100.10%(RSD≤2.12%).CONCLUSION The method is simple,stable and reliable,which can be used for the determination of nucleosides in Dendrobium officinale.
