摘 要:本文旨在探究FABP1基因在猪脂肪沉积性状中所发挥的作用,试验选用180日龄藏猪与大约克猪,对藏猪和大约克猪FABP1基因5’侧翼区(起始密码子上游3000 bp左右序列)进行了单核苷酸多态性(SNPs)位点筛选,并分析其对FABP1基因转录的影响。采用RT-qPCR检测两试验猪种在肝脏、背最长肌和背脂组织中的FABP1基因的mRNA相对表达量。结果显示,在FABP1基因5’侧翼区存在A-1717G、G-1701A、C-1547T、T-1523C、C-1484A、C-1453A、G-1377A、T-1331G共8个突变位点。在藏猪和大约克猪背最长肌、背脂和肝脏组织中FABP1基因的mRNA相对表达量趋势基本一致,FABP1基因在两品种肝脏组织中的表达量最高。FABP1基因在两品种间8个突变的位点的P值均为极显著差异,通过转录因子预测发现在A-1717G、C-1484A、C-1453A以及G-1377A四个位点中均存在与脂肪相关的转录因子出现或消失。综上,FABP1基因可能为调控脂肪沉积的关键基因。The purpose of this paper was to explore the role of FABP1 gene in pig fat deposition traits.The 180-day-old Tibetan pigs and Yorke pigs were selected for the experiment,and gene sequence were screened for single nucleotide polymorphisms(SNPs) and their effects on FABP1gene transcription were analyzed.In this experiment,RT-qPCR was used to detect the relative m RNA expression of FABP1 gene in liver,longissimus dorsi muscle and dorsal fat tissue of the two experimental pig breeds.The results showed that there were 8 mutation sites including A-1717G,G-1701A,C-1547T,T-1523C,C-1484A,C-1453A,G-1377A and T-1331G in the 5'flanking region of FABP1 gene.The relative mRNA expression trends of FABP1 gene in the longissimus dorsi,dorsal fat and liver tissues of Tibetan pigs and York pigs were basically the same,and the expression level of FABP1 gene was the highest in the liver tissues of the two breeds.The P values of the eight mutated loci in the FABP1 gene were highly significantly different between the two species,and the presence or absence of fat-related transcription factors was found in four loci,A-1717G,C-1484A,C-1453A and G-1377A,by transcription factor prediction.In conclusion,FABP1 gene may be a key gene regulating fat deposition and metabolism.
