摘　　要：吡仑帕奈(Perampanel)是日本卫材公司于2012年10月研发的Fycompa品牌。它是目前为止,唯一对AMPA的受体起效的新一代新药,是第一批用于治疗12岁及以上难治性部分性癫痫发作患者的添加药物,是一种AMPA受体拮抗剂,具有高选择性并调节中枢神经系统中的谷氨酸活性。其药物成分复杂,是经化合物库高通量筛选及后续的结构改造优化而开发的,是以AMPA受体为靶点的首个上市药物。临床上可用于单药或联合用药治疗。常见的不良事件包括头晕、嗜睡、头痛、疲劳、共济失调等。Perampanel is a Fycompa brand developed by Japanese Eisai Company in October 2012.So far,it is the only new drug of a new generation that works on the AMPA receptor.It is the first batch of added drugs used to treat patients with refractory partial seizures aged 12 years and above.It is an AMPA receptor antagonist with high selectivity and regulates the activity of glutamate in the central nervous system.Its drug composition is complex.It was developed through high-throughput screening of the compound library and subsequent structural modification and optimization.It is the first drug to be marketed targeting AMPA receptor.Clinically,it can be used for single or combined drug treatment.Common adverse events include dizziness,lethargy,headache,fatigue,ataxia,etc.
