摘  要:本研究考察含三叶鬼针草提取物的眼部产品的安全性与抗皱紧致功效。采用鸡胚绒毛尿囊膜试验(HET-CAM Test)评估三叶鬼针草提取物和三叶鬼针草涂抹眼膜的刺激性与安全性,CCK8法评估三叶鬼针草提取物的细胞毒性,采用斑马鱼模型评估三叶鬼针草涂抹眼膜的抗皱紧致功效,并采用封闭型斑贴试验和人体功效测试进行三叶鬼针草涂抹眼膜的安全性及抗皱紧致功效评估。结果表明三叶鬼针草提取物对眼部具有无/轻刺激性;角质形成细胞(HaCaT)和成纤维细胞(HSF)活性测定发现10^(-4)~10^(-1)g/L的三叶鬼针草提取物对HaCaT无细胞毒性,10^(-4)~10^(-1)g/L的三叶鬼针草提取物对HSF无细胞毒性;含三叶鬼针草的涂抹眼膜可以显著地增加col1a1a、eln1基因相对表达量,对人体皮肤无不良反应,且有显著地降低皱纹面积、提高皮肤弹性紧致度、平滑度的效果,具有显著的抗皱紧致功效。Our objective in this study was to evaluate of the safety and anti-wrinkle and firming effect of the eye mask with Bidens pilosa extract.HET-CAM Test was used to evaluate the irritation and safety of Bidens pilosa extract and the eye mask with Bidens pilosa extract.CCK8 method was used to test the cytotoxicity of the eye mask with Bidens pilosa extract.And zebrafish model was used to evaluate the anti-wrinkle and firming effect of the eye mask with Bidens pilosa extract.The safety and anti-wrinkle and firming efficacy of the eye mask with Bidens pilosa extract were evaluated by patch test and human efficacy test.The results showed Bidens pilosa extract had not irritation or mildly irritation,and keratinocytes(HaCaT)and fibroblast(HSF)activities test revealed that 10^(-4)~10^(-1) g/L Bidens pilosa extract had no cytotoxic on HaCaT and 10^(-4)~10^(-1) g/L Bidens pilosa extract has no cytotoxic on HSF.The relative expression levels of col1a1a and eln1 genes were significantly increased by using the eye mask with Bidens pilosa L.There was no adverse effect on human skin,and significantly improved skin texture and water content(P<0.05),and also improved skin elasticity around the eyes to a certain extent.It had significant anti-wrinkle and firming effect.
