摘 要:目的:探讨地五养肝胶囊(DWYG)对左旋谷氨酸单钠(MSG)大鼠-肝再生模型肝脏损伤后再生修复的影响及其相关机制。方法:MSG大鼠腹腔注射50%四氯化碳(CCl 4)/大豆油,建立MSG大鼠-肝再生模型。同步给予DWYG干预后,采用HE染色和Masson染色检测DWYG对MSG大鼠-肝再生模型肝脏损伤后再生修复反应的影响;采用ELISA试剂盒检测DWYG对MSG大鼠-肝再生模型血清瘦素浓度的影响;应用Western Blot检测DWYG对MSG大鼠-肝再生模型肝脏瘦素受体蛋白表达水平的影响;应用荧光实时定量PCR检测DWYG对MSG大鼠-肝再生模型肝脏Hedgehog信号相关分子mRNA表达水平的影响。结果:HE染色和Masson染色结果显示DWYG干预可以显著减轻MSG大鼠-肝再生模型肝脏组织损伤和纤维化程度;ELISA检测提示DWYG干预可明显降低MSG大鼠-肝再生模型血清瘦素浓度,而Western Blot检测显示DWYG可以显著降低纤维化肝组织中瘦素受体的蛋白表达水平。荧光实时定量PCR检测结果提示DWYG干预可以下调MSG大鼠-肝再生模型纤维化肝组织中Hh信号通路相关分子(Shh、Gli1、Smo和Ptc)的mRNA表达水平。结论:DWYG可以通过干预MSG大鼠-肝再生模型体内异常的瘦素信号从而减轻肝脏损伤后的纤维性再生修复。Objective:To investigate the effects and mechanisms of Diwu Yanggan Capsules on liver regeneration and repair after injury of monosodium glutamate(MSG)-rat-liver regeneration models.Methods:The MSG-rat-liver regeneration model was established in MSG-rats by injecting a 50%CCl 4/soybean oil solution subcutaneously.After DWYG treatment,HE stain and Masson stain were carried out to examine the effects of DWYG on liver injury and fibrosis degree.The impact of DWYG on the serum leptin level of MSG-rat-liver regeneration models was further examined by ELISA.And the impact of DWYG on the protein expression level of hepatic leptin receptor(Ob-Rb)in MSG-rat-liver regeneration models was further examined by Western blotting analysis.Then the impact of DWYG on the mRNA expression level of Hedgehog signal pathway in the liver tissues of MSG-rat-liver regeneration models was further examined by quantitative real-time RT-PCR analysis.Results:The results of HE stain and Masson stain showed that DWYG treatment could ameliorate liver injury and inhibit the collagen accumulation in liver tissues of MSG-rat-liver regeneration models.Moreover,the results of ELISA demonstrated that after DWYG treatment the serum level of leptin in MSG-rat-liver regeneration models was declined.Meanwhile,the down-regulated protein expression level of Ob-Rb in MSG-rat-liver regeneration models after DWYG treatment was verified by Western blotting.Furthermore,the quantitative real-time RT-PCR analysis demonstrated that after DWYG treatment the mRNA expression levels of related molecules of Hh signaling pathway(Shh,Smo,Ptc and Gli1)in liver tissues of MSG-rat-liver regeneration models were dramatically repressed.Conclusion:DWYG could efficiently attenuate the fibrotic repair in the injured liver tissues of MSG-rat-liver regeneration models by intervening in abnormal leptin signaling.
