摘  要:目的研究血水草Eomecon chionantha Hance化学成分及其抗炎活性。方法血水草70%乙醇提取物采用硅胶、Sephadex LH-20、大孔树脂、MIC、制备TLC、半制备HPLC进行分离纯化,根据理化性质及波谱数据鉴定所得化合物的结构。采用RAW264.7细胞评价其体外抗炎活性。结果从中分离得到15个化合物,分别鉴定为6-carboxymethyldihydrochelerythrine(1)、(R)-8-[(R)-1-hydroxyethyl]dihydrochelerythrine(2)、zanthoxyline dimethoxy derivative(3)、oxychelerythrine(4)、6-methyldihydrochelerythrine(5)、macleayin E(6)、8-hydroxydihydrochelerythrine(7)、1-(dihydrosanguinarine-6-yl)ethanol(8)、methyl 2′-(7,8-dihydrosanguinarine-8-yl)acetate(9)、(3β,18β,20β)-3-hydroxy-olean-12-en-29-oic acid(10)、3β,22β-dihydroxy-olean-12-en-29-oic acid(11)、齐墩果酸(12)、β-amyrin(13)、2,2′-oxybis(1,4-di-tert-butylbenzene)(14)、棕榈酸(15)。化合物1~3、10~13具有较好的抑制NO释放的活性,其IC_(50)值分别为(7.77±0.18)、(24.40±0.23)、(5.03±0.15)、(7.17±0.02)、(3.16±0.10)、(7.34±0.03)、(29.73±0.02)μg/mL。结论化合物1~2、4、6、8~12、14~15为首次从该植物中分离得到。化合物1~3、10~13有较好的抗炎活性。AIM To study the chemical constituents from Eomecon chionantha Hance and their anti-inflammatory activities.METHODS The 70%ethanol extract from E.chionantha was isolated and purified by silica gel,Sephadex LH-20,macroporous resin,MIC,preparative TLC and semi-preparative HPLC,then the structures of obtained compounds were identified by physicochemical properties and spectral data.Their anti-inflammatory activities in vitro were evaluated by RAW264.7 cells.RESULTS Fifteen compounds were isolated and identified as 6-carboxymethyldihydrochelerythrine(1),(R)-8-[(R)-1-hydroxyethyl]dihydrochelerythrine(2),zanthoxyline dimethoxy derivative(3),oxychelerythrine(4),6-methyldihydrochelerythrine(5),macleayin E(6),8-hydroxydihydrochelerythrine(7),1-(dihydrosanguinarine-6-yl)ethanol(8),methyl 2′-(7,8-dihydrosanguinarine-8-yl)acetate(9),(3β,18β,20β)-3-hydroxy-olean-12-en-29-oic acid(10),3β,22β-dihydroxy-olean-12-en-29-oic acid(11),oleanolic acid(12),β-amyrin(13),2,2′-oxybis(1,4-di-tert-butylbenzene)(14),palmitic acid(15).Compounds 1-3,10-13 had good activities of inhibiting NO release,and their IC_(50)values were(7.77±0.18),(24.40±0.23),(5.03±0.15),(7.17±0.02),(3.16±0.10),(7.34±0.03),(29.73±0.02)μg/mL,respectively.CONCLUSION Compounds 1-2,4,6,8-12,14-15 are isolated from this plant for the first time.Compounds 1-3,10-13 show good anti-inflammatory activities.
