木芙蓉叶质量标准的提升研究

摘　　要：目的提升木芙蓉叶质量标准。方法在原标准的基础上,优化供试品制备方法,增加银锻苷TLC鉴别,采用UPLC法同时测定芦丁、异槲皮苷、山柰酚-3-O-芸香糖苷、银锻苷的含量。该药材60%甲醇提取液的分析采用Utimate UPLC AQ-C_(18)色谱柱(100 mm×2.1 mm,1.8μm);流动相乙腈-0.1%甲酸,梯度洗脱;体积流量0.25 mL/min;柱温40℃;检测波长260 nm。结果银锻苷TLC特征斑点清晰,分离度好。芦丁、异槲皮苷、山柰酚-3-O-芸香糖苷、银锻苷在各自范围内线性关系良好(r≥0.9996),平均加样回收率为95.26%~102.52%,RSD为1.05%~2.63%。结论该方法准确可行,可用于木芙蓉叶的质量控制。建议木芙蓉叶含芦丁、异槲皮苷、山柰酚-3-O-芸香糖苷、银锻苷总含量不得少于2.1996 mg/g,即0.22%。AIM To improve the quality standard for the leaves from Hibisci Mutabilis Folium.METHODS On the basis of the original standard,the preparation method of the sample was optimized by the TLC identification of tiliroside,and the simultaneous UPLC content determination of rutin,isoquercitrin,kaempferol-3-O-rutin glycoside and tiliroside.The analysis of 60%methanol extract of the herbal medicine was performed on a 40℃thermostatic Utimate UPLC AQ-C_(18) column(100 mm×2.1 mm,1.8μm),with the mobile phase comprising of acetonitrile-0.1%formic acid flowing at 0.25 mL/min in a gradient elution manner,and the detection wavelength was set at 260 nm.RESULTS TLC identification of tiliroside displayed clear and well-separated spots.Rutin,isoquercitrin,kaempferol-3-O-rutin glycoside,tiliroside showed good linear relationships within their own ranges(r≥0.9996),whose average recoveries were 95.26%-102.52%with the RSDs of 1.05%-2.63%.CONCLUSION This accurate and feasible method can be used for the quality control of Hibisci Mutabilis Folium.We suggest no less than 2.1996 mg/g(0.22%)total content of rutin,isoquercitrin,kaempferol-3-O-rutin glycoside and tiliroside in Hibisci Mutabilis Folium.
