摘 要:甜糯山药是西双版纳的特有品种,传统的繁殖方式效率不高。为推广、扩繁该品种,本研究以西双版纳甜糯山药的茎尖和茎段为材料,对各阶段的培养基、培养条件进行筛选优化,获得了一套适合西双版纳甜糯山药茎尖、茎段组培的快繁体系。其中:最适诱导培养基为MS+6-BA 2.0 mg/L+2,4-D 2.0 mg/L+PVP 150 mg/L+VC600 mg/L+蔗糖30 g/L+琼脂7.8 g/L;最适分化培养基为MS+6-BA 2.0 mg/L+NAA 2.0 mg/L+PVP 150 mg/L+VC600 mg/L+蔗糖30 g/L+琼脂7.8 g/L,培养30 d分化率可达到73.33%;最适生根培养基为1/2MS+NAA 0.5 mg/L+6-BA 0.5 mg/L+蔗糖15 g/L+琼脂7.8 g/L,培养30 d后生根率达到80.00%;最后组培苗通过炼苗移栽至装有育苗土(沙∶土∶基质=1∶1∶1)的穴盘中,10~15 d后成活率可达65%以上。Tiannuo yam is unique variety in Xishuangbanna,the efficiency of traditional breeding way is not high.To promote and propagate the variety,this study screened and optimized the culture medium and conditions at each stage by using the stem tips and segments of Xishuangbanna Tiannuo yam as materials.This study obtained a set of rapid propagation system suitable for tissue culture of Xishuangbanna Tiannuo yam stem tips and segments.Among them,the optimal induction medium was MS+6-BA 2.0 mg/L+2,4-D 2.0 mg/L+PVP 150 mg/L+VC 600 mg/L+sucrose30 g/L+agar 7.8 g/L.The optimal differentiation medium was MS+6-BA 2.0 mg/L+NAA 2.0 mg/L+PVP 150 mg/L+VC600 mg/L+sucrose 30 g/L+agar 7.8 g/L,and the differentiation rate could reach 73.33% after cultivation for 30 days.The optimal rooting medium was 1/2MS+NAA 0.5 mg/L+6-BA 0.5 mg/L+sucrose 15 g/L+agar 7.8 g/L,and the rooting rate could reache 80.00% after cultivation for 30 days.Finally,the tissue cultured seedlings were transplanted into the hole tray filled with nursery soil(sand∶soil∶substrate=1∶1∶1) after acclimatization,and the survival rate could reach more than 65% after 10-15 days.
