老年糖尿病肾病患者ABCA1基因多态性与动脉粥样硬化相关性的研究

摘　　要：目的探讨老年糖尿病(DM)患者与糖尿病肾病(DKD)患者中ABCA1 rs2230806(A>G)基因多态性与DKD动脉粥样硬化的相关性。方法利用TaqMan SNP基因分型分析400例老年DM与DKD患者rs2230806基因多态性,进行卡方检验和多变量逻辑回归分析,评估rs2230806与DKD易感性之间的关联。结果主要等位基因(A)与DKD高风险之间存在显著相关性(P=0.011)。在显性模型中,AA较AG+GG基因型的DKD风险显著升高(P=0.019)。在动脉粥样硬化指数(AI)异常组隐性模型中,AA+AG较GG基因型的心血管事件的发生率显著降低(P=0.018);在AI异常组显性模型中,AG+GG较AA基因型的心血管事件的发生率显著降低(P=0.047)。结论ABCA1 rs2230806多态性A等位基因与DKD发病风险及心血管事件的发生率具有正相关性。Objective To investigate the correlation between polymorphisms of ABCA1 rs2230806(A>G)and atherosclerosis(AS)in elderly patients diabetes mellitus(DM)and diabetic kidney disease(DKD).Methods The polymorphisms of ABCA1 rs2230806 were analyzed by using TaqMan SNP genotyping in DM patients and DKD patients(n=400),and given chi-square test and multivariate Logistic regression analysis.The correlation between rs2230806 and DKD susceptibility was reviewed.Results There was a significant correlation between major allele(A)and DKD high risk(P=0.011).In dominant model,DKD risk of genotype AA was significantly higher than that of genotype AG+GG(P-0.019).The incidence rates of cardiovascular events were significantly lower in genotype AA+AG than those in genotype GG in recessive model of group of abnormal atherosclerosis index(AI,P=0.018).The incidence rates of cardiovascular events were significantly lower in genotype AA+AG than those in genotype AA in dominant model of abnormal AI group(P=0.047).Conclusion There is a positive correlation among allele A of ABCA1 rs2230806 polymorphisms,DKD risk and incidence rates of cardiovascular events.
