脱钙液的组分与处理时间对骨髓活检组织HE和免疫组化染色的影响

摘　　要：目的:探讨不同组分的脱钙液及脱钙时间对骨髓活检组织HE染色和免疫组化染色效果的影响。方法:收集36例骨髓活检穿刺组织,分为10%硝酸甲醛液(A液)、10%盐酸甲醛液(B液)和30%甲酸甲醛液(C液)3组,每组12例,分别观察各组的脱钙时间。另收集5例骨髓活检穿刺组织,将同一例标本分成两份,观察不同组分的脱钙液和脱钙处理时间对HE染色和免疫组化染色效果的影响。结果:A液、B液的脱钙时间比C液短;3种方法的HE染色效果均较好;而C液脱钙2 h的Ki67免疫组化染色效果好,但其他方法的Ki67免疫组化染色效果欠佳。结论:采用30%甲酸甲醛液处理2 h的脱钙法,实验操作简便,HE染色和免疫组化染色效果较为满意。Objective:To investigate the effects of different components of decalcification solutions and decalcification time on HE and immunohistochemical staining of bone marrow biopsy tissues.Methods:36 cases of bone marrow biopsies were collected,which were divided into three groups,with 12 cases in each group.Decalcification time of 10% nitric acid formaldehyde solution(solution A),10% hydrochloric acid formaldehyde solution(solution B) and 30% formic acid formaldehyde solution(solution C) was observed respectively.Another five bone marrow biopsies were collected and the same sample was divided into two parts,whose effects of different decalcification solutions and decalcification time on HE and immunohistochemical staining were observed.Results:The decalcification time of solution A and solution B was shorter than that of solution C.The HE staining effects decalcified by all methods were good.The Ki67 immunohistochemical staining effect was even better when the sample was decalcified with the 30% formic acid formaldehyde solution for 2 h.And the Ki67 immunohistochemical staining effects decalcified by other methods were not good enough.Conclusion:Decalcification with 30% formic acid formaldehyde solution for 2 h is simple and convenient,and the effects of HE and immunohistochemical staining are satisfactory.
