摘 要:目的优化白扁豆、瓜蒌子药材粉末饮片的煎煮工艺。方法采用高效液相色谱(HPLC)法检测白扁豆、瓜蒌子药材粉末饮片中核苷类成分的含量。以核苷类成分总含量为评价指标,以粉碎粒度、提取时间、提取次数为考察因素,优化煎煮工艺,并验证。结果胞嘧啶、尿嘧啶、尿苷、鸟苷、腺苷进样量分别在0.0342~0.3423μg,0.0375~0.4569μg,0.0383~0.3831μg,0.0415~0.4146μg,0.0454~0.4538μg范围内与峰面积线性关系良好(r=1.0000);精密度、稳定性、重复性试验结果的RSD均小于2.0%;白扁豆药材粉末饮片样品中胞嘧啶、尿嘧啶、尿苷、腺苷平均加样回收率分别为97.55%,101.25%,98.87%,99.46%,RSD分别为1.84%,2.62%,2.22%,1.86%(n=9);瓜蒌子药材粉末饮片样品中胞嘧啶、尿嘧啶、尿苷、鸟苷、腺苷平均加样回收率分别为98.72%,100.80%,99.16%,99.26%,103.30%,RSD分别为4.84%,4.46%,2.12%,6.11%,4.03%(n=9)。白扁豆、瓜蒌子药材粉末饮片的最优煎煮工艺为,将药材饮片打成粗粉(后者需过2号筛),煎煮2次,每次20 min。结论本研究中优化所得工艺可为药材粉末饮片煎煮的规范化研究提供参考。Objective To optimize the decoction process of Lablab Semen Album and Trichosanthis Semen Powder Decoction Pieces.Methods The content of nucleosides in the Lablab Semen Album and Trichosanthis Semen Powder Decoction Pieces was determined by the high-performance liquid chromatography(HPLC)method.The decoction process was optimized with the total content of nucleosides as the evaluation indicator,and with the crushing granule size,extraction time and extraction frequency as the investigation factors.Then,the verification test was carried out.Results The linear ranges of cytosine,uracil,uridine,guanosine and adenosine were 0.0342-0.3423µg,0.0375-0.4569µg,0.0383-0.3831µg,0.0415-0.4146µg and 0.0454-0.4538µg respectively(r=1.0000).The RSDs of precision,stability and repeatability tests were all lower than 2.0%.The average recoveries of cytosine,uracil,uridine and adenosine in the Lablab Semen Album Powder Decoction Pieces were 97.55%,101.25%,98.87%,99.46%,with RSDs of 1.84%,2.62%,2.22%,1.86%respectively(n=9).The average recoveries of cytosine,uracil,uridine,guanosine and adenosine in the Trichosanthis Semen Powder Decoction Pieces were 98.72%,100.80%,99.16%,99.26%,103.30%,with RSDs of 4.84%,4.46%,2.12%,6.11%,4.03%respectively(n=9).The optimal decoction process of Lablab Semen Album and Trichosanthis Semen Powder Decoction Pieces was as follows:crushing the medicinal decoction pieces into the coarse powder(the coarse powder of Trichosanthis Semen needing to pass through a No.2 sieve),decocting twice,each time for 20 min.Conclusion The optimal process can provide a reference for the standardization of decoction of medicinal powder decoction pieces.
