摘 要:目的:基于网络药理学方法分析阐释参麻益智方的配伍规律及其治疗血管性痴呆(VaD)的合理性。方法:对参麻益智方中4味中药人参、鬼箭羽、天麻、川芎、全方及不同配伍的预测靶点进行京都基因和基因组百科全书(KEGG)信号通路富集分析,构建“成分-靶点”网络等,发现每味中药及全方可能参与的信号通路。通过STRING网站构建参麻益智方靶点和VaD发病共同靶点的蛋白-蛋白互作(PPI)网络,并进行KEGG信号通路富集分析,发现参麻益智方治疗VaD的关键干预靶点及可能的作用机制。结果:参麻益智方的预测靶点共291个。KEGG信号通路富集分析发现每味中药及全方都参与代谢及血管功能调节。人参及天麻的共同作用靶点93个,人参及川芎的共同作用靶点91个,天麻和川芎可协助君药人参改善机体血管功能。参麻益智方靶点和VaD发病的共同靶点主要包括白细胞介素6、肿瘤坏死因子、载脂蛋白E、半胱氨酸蛋白酶3及脑源性神经营养因子等。KEGG信号通路富集分析显示参麻益智方治疗VaD的主要调控部位在血管,与改善脑内血管炎症有关。结论:参麻益智方配伍严谨、合理,其治疗VaD可能与改善脑血管内皮细胞炎症有关。Objective:To analyze the compatibility and rationality of Shenma Yizhi Recipe in the treatment of vascular dementia(VaD) based on network pharmacology.Methods:The Kyoto Encyclopedia of Genes and Genomes(KEGG) signaling pathway enrichment analysis were performed on the predicted targets of the four herbs,the whole formula,and the different combinations of the formula,and the "component-target" network was constructed to find out the possible signaling pathways that might be involved in individual herbs and the whole recipe.The protein-protein interactions(PPI) network of the targets of Shenma Yizhi Recipe and the common targets of VaD were constructed through the STRING website,and KEGG signaling pathway enrichment analysis was performed to find out the key interventional targets of Shenma Yizhi Recipe for the treatment of VaD and the possible mechanism of action.Results:There were 291 predictive targets of Shenma Yizhi Recipe,and KEGG signal pathway enrichment showed that individual Chinese herb and the whole recipe were all involved in the regulation of metabolism and vascular function.There were 93 common targets of Panax Ginseng and Euonymus Alatus,and 91 common targets of Panax Ginseng and Ligusticum Wallichii.Euonymus Alatus and Ligusticum Wallichii could assist Panax Ginseng to improve vascular function.The main common targets of Shenma Yizhi Recipe and VaD pathogenesis included interleukin 6,tumor necrosis factor,apolipoprotein E,Caspase-3,and brain-derived neurotrophic factor.KEGG signaling pathway enrichment analysis showed that the main regulatory site of Shenma Yizhi Recipe in treating VaD was blood vessels,which was related to improve vascular inflammation in the brain.Conclusion:The compatibility of Shenma Yizhi Recipe was rigorous and rational,and its treatment of VaD might be related to the amelioration of cerebrovascular endothelial inflammation.
