摘  要:目的探讨不同时期川崎病(KD)患儿血浆中hsa_circ_0094885的表达水平及是否可作为KD的潜在诊断生物标志物。方法选取确诊KD患儿23例,再选取同期同年龄段20例感染患儿和20例非感染患儿为对照组,通过qRT-PCR技术检测所有患儿circRNA血浆中的表达水平,比较各组间circRNA表达差异性和构建受试者操作特征曲线。结果KD急性期血浆hsa_circ_0094885表达水平低于感染组及非感染组(P<0.05),KD恢复期血浆hsa_circ_0094885表达水平高于急性期(P<0.05),KD急性期血浆hsa_circ_0094885的ROC曲线下面积为0.763,标准误0.072,95%置信区间0.621~0.905,截断值为0.46,敏感度为78.26%,特异度为78.57%。结论血浆hsa_circ_0094885可作为急性期KD新型诊断生物标志物,血浆hsa_circ_0094885可能参与KD的发病机制,可作为KD转归的评价指标。Objective To explore the plasma levels of hsa_circ_0094885 in children with Kawasaki disease(KD)at different stages and whether it can be used as a potential diagnostic Biomarker of KD.Methods Twenty-three children diagnosed KD were selected as the study group,and 20 infected children and 20 non infected children of the same age group were selected as the control group.The expression level of circRNA in the plasma of the study subjects was detected by qRT-PCR,and the difference of circRNA expression between the groups was compared,the ROC was constructed for evaluating the diagnostic value of hsa_circ_0094885 in plasma of acute stage of Kawasaki disease.Results The expression level of hsa_circ_0094885 in plasma of acute stage of Kawasaki disease patients were significantly lower than that in the control groups(P<0.05).The expression level of hsa_circ_0094885 in plasma of convalescent stage of Kawasaki disease was significantly higher than in the acute stage patients(P<0.05).The area under ROC curve of hsa_circ_0094885 in acute Kawasaki disease plasma was 0.763,the standard error was 0.072,the 95%confidence interval was 0.621~0.905,the cut-off value was 0.46,the sensitivity was 78.26%,and the specificity was 78.57%.Conclusion Plasma hsa_circ_0094885 can be used as a new diagnostic biomarker for acute KD,which may be involved in the pathogenesis of KD and can be used as an evaluation index for the prognosis of KD.
