摘 要:研究以岛津Column Shim-pack GIST C18(250 nm×4.6 nm)为色谱柱,甲醇-水为流动相,流速1 mL/min,柱温40℃,检测样品中的白术内酯。白术内酯Ⅰ和Ⅱ、Ⅲ的检测波长分别为275和220 nm,进样量为10μL,白术内酯Ⅰ的线性关系方程为Y=1.049×10~7X-4.090×10~5(r=0.999 8),在4~1 023μg范围内线性关系良好;白术内酯Ⅱ的线性关系方程为Y=6.872×10~6X+1.194×10~6(r=0.999 0),在3.7~933μg范围内线性关系良好;白术内酯Ⅲ的线性关系方程为Y=4.819×10~6X-3.969×10~5(r=0.999 0),在3.7~933μg范围内线性关系良好。研究还测定了不同产地(山东、安徽、湖南)、不同采收时间(2022年12月1、10、20、27日)的白术和不同温度(50、60、70、80和90℃)所制白术的白术内酯(Ⅰ、Ⅱ、Ⅲ)含量。结果表明:安徽亳州的白术样品中白术内酯Ⅰ、Ⅱ、Ⅲ的含量较高,白术内酯Ⅰ、Ⅱ、Ⅲ含量分别为1.07、0.43、2.40 mg/g;以12月20日采收的白术样品中白术内酯Ⅰ、Ⅱ、Ⅲ的含量较高,分别为1.49、0.53、1.96 mg/g;而50℃烘干白术样品中白术内酯Ⅰ、Ⅱ、Ⅲ的含量较高,分别为1.50、2.37、1.45 mg/g。This study has revealed the differences of contents of atractylenolide Ⅰ,Ⅱ and Ⅲ in Atractylodes macrocephala samples from various origins(Shandong,Anhui,Hunan),harvest time(2022,Dec.1st,10th,20th,and 27th) and drying temperatures(50,60,70,80 and 90℃).The seperation and detection of atractylenolide Ⅰ,Ⅱ and Ⅲ was performed under the conditions of Shimadzu Column Shim-pack GIST C18(250 nm×4.6 nm) chromatographic column,methanol-water mobile phase,1mL/min flow rate,and 40 °C column temperature.The detection wavelength for atractylenolide I was 275 nm,for atractylenolide Ⅱ and Ⅲ was 220 nm,and the injection volume was 10μL.The linear equation of atractylenolide Ⅰ was Y=1.049×10~7X-4.090 ×10~5(r=0.999 8),performing well in the range of 4-1,023 μg.The linear equation of atractylenolide Ⅱ was Y=6.872×10~6X+1.194×10~6(r=0.999 0),performing well in the range of 3.7-933 μg.The linear equation of atractylenolide Ⅲ was Y=4.819×10~6X-3.969×10~5(r=0.999 0),acting well in the range of 3.7-933 μg.The detection results showed that the contents of atractylenolide Ⅰ,Ⅱ and Ⅲ in the samples from Bozhou,Anhui were high,reaching 1.07,0.43 and 2.40 mg/g,respectively,in the samples harvested on Dec.20th were high,reaching 1.49,0.53 and 1.96 mg/g,respectively,and in the samples dried at 50℃ were high,reaching 1.50,2.37 and 1.45 mg/g,respectively.
