摘  要:目的研究夏枯草乙酸乙酯提取物对人脐静脉内皮细胞融合细胞(EA·hy926)中内皮型一氧化氮合酶(eNOS)的作用。方法体外培养EA·hy926,将其分为夏枯草乙酸乙酯提取物高、中、低剂量组和阴性对照组,分别用31.20、15.60、7.80μg/mL夏枯草乙酸乙酯提取物和5‰DMSO的培养基孵育24 h。通过试剂盒检测一氧化氮(NO)和eNOS含量。采用Western blot检测eNOS、磷酸化内皮型一氧化氮合酶(p-eNOS)、热激蛋白90(HSP-90)、窖蛋白-1(Cav-1)的表达。结果试剂盒结果显示中、高剂量组细胞内NO和eNOS含量高于阴性对照组,差异有统计学意义(P<0.05)。Western blot结果显示中、高剂量组eNOS和HSP-90的蛋白表达高于阴性对照组,Cav-1的蛋白表达低于阴性对照组,差异有统计学意义(P<0.05);高剂量组p-eNOS的蛋白表达高于阴性对照组,差异有统计学意义(P<0.05)。结论夏枯草乙酸乙酯提取物通过增加血管内皮细胞内eNOS、p-eNOS的表达,增加NO释放;此外,其还能提高HSP-90的蛋白表达,降低Cav-1的蛋白表达,促进eNOS释放NO,从而舒张血管,降低血压。Objective To study the effects of ethyl acetate extract of prunella vulgaris on endothelial nitric oxide synthase(eNOS)synthesis by human umbilical vein endothelial cells(EA·hy926).Methods EA·hy926 were cultured in vitro and divided into high,medium and low dose groups of prunella vulgaris ethyl acetate extract and negative control group,31.20,15.60,7.80μg/mL ethyl acetate extract of prunella vulgaris and 5‰DMSO were incubated for 24 hours.Detect the content of nitric oxide(NO)and eNOS through the kit.The expressions of eNOS,phosphorylated endothelial nitric oxide synthase(p-eNOS),heat shock proteins 90(HSP-90)and caveolin-1(Cav-1)were detected by Western blot.Results The results of the kit showed that the contents of intracellular NO and eNOS in the medium and high dose groups were higher than those in the negative control group,and the difference was statistically significant(P<0.05).Western blot results showed that the protein expression of eNOS and HSP-90 in the medium and high dose groups was higher than that in the negative control group,and the protein expression of Cav-1 was lower than that in the negative control group,the difference was statistically significant(P<0.05);the protein expression of p-eNOS in the high dose group was higher than that in the negative control group,the difference was statistically significant(P<0.05).Conclusion The ethyl acetate extract of prunella vulgaris increased NO release by increasing the expression of eNOS and p-eNOS in vascular endothelial cells;in addition,it can also increase the protein expression of HSP-90,reduce the protein expression of Cav-1,promote eNOS to release NO,so as to relax blood vessels and reduce blood pressure.
