摘 要:目的研究黄芩汤中不同药物与黄芩配伍对君药黄芩有效成分含量和抗皮肤癣菌活性的影响,并探讨两者的相关性。方法采用L8(27)正交设计法组方,以高效液相色谱法(HPLC)测定各配伍样品中黄芩苷、汉黄芩苷、黄芩素、汉黄芩素、千层纸素A、千层纸素A苷6种有效成分的含量;以临床常见皮肤癣菌——须癣毛癣菌为例,采用微量稀释法测定黄芩不同配伍组的最小抑菌浓度(MIC值),比较配伍前后的抗真菌作用;采用Pearson分析两者的相关性。结果白芍、甘草和大枣在用药水平时,黄芩苷、黄芩素等各成分含量均大于不用药水平,其对成分含量的影响程度为:白芍>甘草>大枣;白芍的用药作用对各成分含量的影响均差异有统计学意义(P<0.05);四味药合煎时各成分含量最高。白芍、甘草在用药水平时,汤液MIC值小于不用药水平,而大枣在用药水平时,其MIC值大于不用药水平;四味药合煎液MIC值最小。Pearson相关分析显示黄芩苷等6个成分与MIC值均呈显著负相关(r>0.7且P<0.05)。结论黄芩汤的抗皮肤癣菌作用强度与君药黄芩的主要成分含量显著相关,黄芩汤配伍能够促进成分溶出和提高抗真菌活性。Objective To study the effects of compatibility of different drugs in huangqin decoction with scutellariae radix on the content changes of main components of scutellariae radix and anti-dermatophyte activity,and to explore the correlation between components contents and anti-dermatophyte activity.Methods Different prescriptions was composed by L8(27)orthogonal design,the contents of baicalin,oroxylin A 7-O-glucuronide,wogonoside,baicalein,wogonin and oroxylin A in each prescription sample were determined by HPLC.The minimum inhibitory concentrations(MIC)of different prescriptions were determined by microdilution method,taking clinical common dermatophyte trichophyton mentagrophytes as an example,and then the antifungal effects of different prescriptions were compared.The correlation between components contents and anti-dermatophyte activity was analyzed by Pearson correlation analysis.Results The contents of baicalin,baicalein,etc.were higher in compatibility with paeoniae radix alba,glycyrrhrizae radix et rhizoma or jujubae fructus compared with that of non-compatibility group.The degree of influence on the content of components was:paeoniae radix alba>glycyrrhrizae radix et rhizoma>jujubae fructus.The compatibility of paeoniae radix alba had significant influence on the content of components(P<0.05).The content of components was the highest when the four herbs were decocted together.When scutellariae radix was compatibility with paeoniae radix alba or glycyrrhrizae radix et rhizoma,the MIC value of the decoction was lower than that of the non-compatible group,while compatibility with jujubae fructus,the MIC value was higher than that of the non-compatible group.The MIC value of the decoction was the lowest when the four herbs were decocted together.The results of Pearson correlation analysis showed that six components such as baicalin were negatively correlated with MIC values(r>0.7,P<0.05).Conclusion The anti-dermatophyte activity of huangqin decoction was significantly related to the content of main component
